黑果腺肋花楸有效物质的提取及抗氧化和降糖活性研究 被引量:5

Extraction of Effective Substances from Aronia melanocarpa and Their Antioxidant and Hypoglycemic Activities

摘要 以黑果腺肋花楸多酚含量、黄酮含量及出膏率为考察指标,采用单因素实验和正交实验并通过多指标综合评分法优化黑果腺肋花楸多酚和黄酮的提取工艺;通过对DPPH的清除能力评价其抗氧化活性;并通过建立肝细胞高糖损伤模型,比较提取物组与模型组葡萄糖消耗量评价其降糖活性。结果表明,黑果腺肋花楸多酚和黄酮的最佳提取工艺为:超声功率120 W、提取时间10 min、乙醇体积分数50%、液料比10∶1(mL∶g)、提取温度40℃;黑果腺肋花楸提取物的IC 50值(60.350μg·mL^(-)1)与VC的相近,具有较强的抗氧化活性;黑果腺肋花楸提取物溶液(1000~3.125μg·mL^(-)1)无肝细胞毒性,且与模型组相比,提取物组葡萄糖消耗量显著提升,由5.59 mmol·L^(-)1·(24 h)^(-1)升至8.29 mmol·L^(-)1·(24 h)^(-1),具有较强的降糖活性。 Using polyphenols content,flavonoids content,and extraction rate of solid extracts as indexes,we optimized the extraction process of polyphenols and flavonoids from Aronia melanocarpa by single-factor experiments,orthogonal experiments,and multi-index comprehensive scoring method,and evaluated the antioxidant activity by DPPH scavenging ability.Moreover,through establishing a high glucose-injured hepatocyte model,we evaluated the hypoglycemic activity by comparing the glucose consumption between the extract group and the model group.The results show that the optimal extraction conditions of polyphenols and flavonoids are determined as follows:the ultrasonic power of 120 W,the extraction time of 10 min,the ethanol volume fraction of 50%,the liquid-solid ratio of 10∶1(mL∶g),and the extraction temperature of 40℃.The IC 50 value(60.350μg·mL^(-1))of the Aronia melanocarpa extracts is similar to that of VC,indicating that the Aronia melanocarpa extracts have strong antioxidant activity.The Aronia melanocarpa extracts solution(1000~3.125μg·mL^(-)1)has no hepatotoxicity,and the glucose consumption of the extract group is significantly increased from 5.59 mmol·L^(-)1·(24 h)^(-1) to 8.29 mmol·L^(-)1·(24 h)^(-1) compared with that of the model group,indicating that the Aronia melanocarpa extracts have strong hypoglycemic activity.
