高山被孢霉产孢培养基的筛选 被引量:1

Screening of sporulation medium for Mortierella alpina

摘要 为了促进高山被孢霉产孢,研究分析了8种常用于促进丝状真菌产孢的培养基对高山被孢霉菌丝生长和孢子产量的影响,并对产孢培养基进行了改良。结果表明,在40 g/L的玉米粉培养基中补加10 g/L葡萄糖和无机盐(硝酸盐(2 g/L KNO_(3))、磷酸盐(1 g/L NaH_(2)PO_(4))与镁离子(0.3 g/L MgSO_(4)·7H_(2)O),接种高山被孢霉后于28℃培养10 d,再在4℃培养14 d,孢子平均产量达到8.57×10^(6)个/mL,是目前用于高山被孢霉菌株保藏的葡萄糖酵母培养基(GY)(孢子平均产量3.85×10^(6)个/mL)的2.23倍。因此,在有少量葡萄糖和无机盐促进菌丝生长的前提下,40 g/L的玉米粉最适合高山被孢霉产孢。 In order to promote the sporulation of Moritierella alpina, the effects of eight mediums commonly used to promote the sporulation of filamentous fungi on Mortierella alpina mycelium growth and spore production were analyzed, and the sporulation medium was improved. The results showed that the average spore production in 40 g/L corn flour medium supplemented with 10 g/L glucose and inorganic salts(nitrate(2 g/L KNO_(3)), phosphate(1 g/L NaH_(2)PO_(4)) and magnesium ion(0.3 g/L MgSO_(4)·7 H_(2)O) after incubation at 28 ℃ for 10 d and then at 4 ℃ for 14 d reached 8.57×10^(6) spores/mL, which was 2.23 times as high as that in the glucose yeast medium(GY)(3.85×10^(6) spores/mL) currently used for the preservation of Mortierella alpina strains. Therefore, 40 g/L of corn flour was the most suitable for the sporulation of Mortierella alpina in the presence of a small amount of glucose and inorganic salts to promote mycelium growth.
