摘要
目的观察云克对骨质疏松症患者成骨功能的影响。方法取骨质疏松症患者骨组织,酶消化法培养成骨细胞,给予不同浓度云克予以干预,CCK-8法检测细胞增殖功能、流式细胞仪检测细胞周期、对硝基苯磷酸盐法检测碱性磷酸酶(ALP)活性、茜素红染色计数矿化结节的变化、Real-time RT-PCR检测骨钙素和骨形成蛋白(BMP)-2基因的表达。结果在10-9 mol/L~10-5 mol/L浓度条件下云克对成骨细胞均有促进增殖的作用(P<0.05);在10-8 mol/L浓度云克作用下,成骨细胞S期细胞均明显增多(P<0.05),G0/G1期细胞明显减少(P<0.05),细胞增殖指数(PI)明显增高(P<0.05);在10-9 mol/L~10-5 mol/L组浓度条件下云克对成骨细胞ALP活性均有促进作用(P<0.05);在10-8mol/L浓度云克作用下,成骨细胞形成矿化结节数量增多(P<0.05),BMP-2 mRNA表达明显增高(P<0.05),骨钙素mRNA表达稍有增高,但差异无统计学意义(P>0.05)。结论在一定浓度条件下云克具有促进骨质疏松症患者成骨功能的作用。
Objective To observe the effect of Yunke on osteogenic function in patients with osteoporosis.Methods Osteoblasts were collected from osteoporotic patients and cultured after enzyme digestion.They were treated with different concentrations of Yunke.Cell proliferation was assessed with CCK-8 kit.Cell cycle was assessed with flow cytometry.Alkaline phosphatase was assessed with p-nitrophenyl phosphate method.The mineralized nodule was counted with Alizarin red staining.The mRNA expression of osteocalcin and BMP-2 was detected with RT-PCR.Results YunKe increased the proliferation of osteoblasts at the concentrations from 10-9 mol/L to 10-5 mol/L(P<0.05).At the concentration of 10-8 mol/L YunKe,the S phase cells and cell proliferation index(PI)of osteoblasts increased(P<0.05),and the G0/G1 cells reduced.ALP activity of osteoblasts increased with Yunke at concentrations from 10-9 mol/L to 10-5 mol/L(P<0.05).The number of mineralized nodules was the most with Yunke at concentration of 10-8 mol/L(P<0.05).The expression of BMP-2 mRNA significantly increased(P<0.05),and the expression of osteocalcin mRNA slightly increased(P>0.05).Conclusion Yunke promotes the osteogenic function of patients with osteoporosis within a certain concentration range.
