去氢骆驼蓬碱对甲状腺未分化癌患者的细胞放疗增敏作用及分子机制 被引量:1

摘要 目的探讨去氢骆驼蓬碱(HM)对甲状腺未分化癌患者的细胞放疗增敏作用及其分子机制。方法将甲状腺未分化癌细胞分为单独放疗组与HM联合放疗组。两组均转染HIF-1αmrRNA;采用MTT法检测细胞增殖,流式细胞术检测细胞凋亡,采用RT-PCR和Western blot检测两组细胞的HIF-α、PI3K、AKT、MAPK、GLUT、VEGF蛋白表达情况,以此来检测放疗的敏感性。结果HM联合放疗组细胞中HIF-α、PI3K、AKT、MAPK、GLUT、VEGF蛋白及存活率降低,凋亡率升高,放疗敏感性增加;HM可显著抑制HIF-1α蛋白的表达,且该抑制作用具有浓度依赖性。结论在放疗过程中,与HM联用能够对放疗起到增敏作用,提高对甲状腺未分化癌细胞的毒性。 Objective To investigate and analyze the sensitization effect of harmaine(HM)on radiotherapy of anaplastic thyroid cancer cells and its molecular mechanism.Methods The undifferentiated thyroid cancer were divided into radiotherapy alone group and HM combined radiotherapy group.Both groups were transfected with HIF-1αmrRNA.Determined by MTT method was used to detect cell proliferation and flow cytometry to detect cell apoptosis.By RT-PCR and Western blot detection of two groups of cells HIF-alpha,PI3K,and AKT,MAPK,GLUT,the expression of VEGF protein to detect radiation sensitivity.Results HM combined radiotherapy group of cells of HIF-alpha,PI3K,and AKT,MAPK,GLUT,VEGF protein and reduced survival rate,apoptosis rate increased,increased,the radiation sensitivity;HM can significantly inhibit the expression of HIF-1 alpha protein,and this inhibition has the concentration dependence.Conclusion In the process of radiotherapy,the combined use of HM can enhance the sensitization of radiotherapy and increase the toxicity to undifferentiated thyroid cancer cells.
