膈下逐瘀汤加减方对气滞血瘀型慢性输卵管炎模型大鼠TLR4、TNF-α表达影响

Effect of Gexia Zhuyu Decoction on the Expression of TLR4 and TNF-α in Chronic Salpingitis Model Rats with Qi Stagnation and Blood Stasis Type

目的探究膈下逐瘀汤加减方对气滞血瘀型慢性输卵管炎模型大鼠输卵管中TLR4、TNF-α因子表达的影响。方法将52只雌性SD大鼠通过随机方法分为4组,分别是空白组(K组)、模型组(M组)、对照组(D组)、膈下逐瘀汤加减方组(G组)各13只。随后将除K组外3组大鼠造成气滞血瘀型慢性输卵管炎大鼠模型,并于造模第5周开始,给予G组膈下逐瘀汤加减方灌胃,同时给予D组康妇炎胶囊悬液灌胃,M组则以同体积纯净水给予灌胃,疗程共28 d。于最后一次给药并禁食12 h后取双侧输卵管,以RT-PCR及Western Blot的方法分别检测输卵管中因子TLR4、TNF-αmRNA及蛋白的表达。结果膈下逐瘀汤加减方可有效降低输卵管中TLR4、TNF-αmRNA因子及蛋白的表达,且明显优于对照组(P<0.01)。结论膈下逐瘀汤加减方可有效调节模型大鼠输卵管中因子TLR4、TNF-αmRNA及蛋白的表达。

Objective To explore the effect of Gexia Zhuyu Decoction on the expression of TLR4 and TNF-αfactors in the fallopian tubes of a rat model of chronic salpingitis with qi stagnation and blood stasis.Methods 52 female SD rats were randomly divided into 4 groups:blank group(K group),model group(M group),control group(D group),Gexia Zhuyu decoction modified group(G Group) 13 each.Subsequently,three groups of rats except group K were made into rat models of chronic salpingitis with stagnation of qi and blood stasis,and group G was given Gexia Zhuyu Decoction Modified and Subtracted Recipe by gavage at the beginning of the fifth week of model building,and group D was given Kang Fuyan capsule suspension was given by gavage,and the M group was given the same volume of purified water by gavage for a total of 28 days.After the last administration and fasting for 12 hours,the oviducts were taken from both sides,and the expressions of the factors TLR4,TNF-α mRNA and protein in the oviducts were detected by RT-PCR and Western Blot methods.Results Gexia Zhuyu Decoction can effectively reduce the expression of factor TLR4,TNF-αmRNA and protein in the fallopian tube,and it is significantly better than the control group(P <0.01).Conclusion Gexia Zhuyu Decoction can effectively regulate the expression of factor TLR4,TNF-αmRNA and protein in the fallopian tubes of model rats,and has a significant therapeutic effect on chronic salpingitis(qi stagnation and blood stasis type).
