ADAR1与食管鳞状细胞癌病理特征及预后的关系

Relationship Between ADAR1 and Pathological Features and Prognosis of Esophageal Squamous Cell Carcinoma

目的探讨作用于RNA的腺苷脱氨酶1(ADAR1)表达水平与食管鳞状细胞癌病理特征及预后的关系。方法收集南阳市第一人民医院2018年1—12月食管鳞癌手术切除的36例新鲜组织标本,应用RT-qPCR和免疫组织化学法检测并比较食管鳞癌及癌旁正常组织中ADAR1表达水平。收集南阳市第一人民医院2014年1月至2016年1月食管鳞癌病理标本102例,应用免疫组织化学法检测癌组织中ADAR1蛋白表达情况,分析其与患者临床病理特征及预后的关系。结果ADAR1在癌组织中表达水平较癌旁正常组织高(P<0.05),ADAR1表达水平与肿瘤浸润深度和淋巴结转移有关(P<0.05)。ADAR1高表达者中位总生存期短于正常表达者(25个月比39个月,P<0.01)。COX比例风险回归模型显示ADAR1表达水平和肿瘤浸润深度是食管鳞癌预后的独立危险因素(P<0.05)。结论ADAR1与食管鳞癌病理特征及预后密切相关,可能参与食管鳞癌的发生发展。

Objective To investigate the relationship between the expression level of adenosine deaminases acting on RNA 1(ADAR1)and the pathological characteristics and prognosis of esophageal squamous cell carcinoma.Methods A total of 36 fresh tissue specimens of esophageal squamous cell carcinoma were collected from Nanyang First People’s Hospital from January to December 2018,and RT-qPCR and immunohistochemistry were used to detect and compare the expression level of ADAR1 in esophageal squamous cell carcinoma and adjacent normal tissues.A total of 102 pathological specimens of esophageal squamous cell carcinoma from January 2014 to January 2016 in Nanyang First People’s Hospital were collected.Immunohistochemistry was used to detect the expression of ADAR1 protein in cancer tissues and its relationship with the clinical pathological characteristics and prognosis of patients was analyzed.Results The expression level of ADAR1 in cancer tissues was higher than that in normal tissues adjacent to cancer(P<0.05).The expression level of ADAR1 was related to the depth of tumor invasion and lymph node metastasis(P<0.05).The median overall survival of patients with high ADAR1 expression level was shorter than those with normal expression level(25 months vs.39 months,P<0.01).The COX proportional hazard regression model showed that the expression level of ADAR1 and the depth of tumor invasion were independent risk factors for the prognosis of esophageal squamous cell carcinoma(P<0.05).Conclusion ADAR1 is closely related to the pathological characteristics and prognosis of esophageal squamous cell carcinoma,and may be involved in the occurrence and development of esophageal squamous cell carcinoma.
