摘要
目的通过网络药理学探究苇茎汤治疗哮喘急性发作的潜在作用机制。方法使用TCMSP数据库筛选苇茎汤药物活性成分,使用Uniprot及TCMSP数据库查找药物活性成分及潜在靶点,运用Cytoscape构建"中药-活性成分-靶点"网络图。在Drug Bank、Gene Cards、TTD等数据库中搜索哮喘急性发作的疾病靶点,将苇茎汤药物活性靶点与哮喘急性发作的疾病靶点取交集,并制作韦恩图。借助String数据库及Cytoscape软件分析交集靶点的蛋白与蛋白相互作用,并绘制PPI图。运用的Cytoscape的MCODE及cytohubba插件探索药物-疾病交集靶点关系。运用Metascape分析平台探索交集靶点的GO、KEGG富集情况,使用在线绘图平台绘制GO富集分析图及KEGG动态气泡图。结果苇茎汤共有22个活性成分,获得54个药物活性靶点。搜索哮喘急性发作靶点827个,将药物-疾病靶点取交集,共有20个交集靶点,其中JUN、CHRM1、CASP8、PTGS2、TGFB1等为核心交集靶点。GO富集分析结果为:血液循环、调节离子传输、乙酰胆碱受体活性、蛋白质结构域特异性结合等。KEGG富集分析结果显示苇茎汤治疗哮喘急性发作主要与组织生长因子(TNF)、核转录因子κB(NF-κB)、神经受体-配体相互作用、PI3K-Akt、钙离子、NOD样受体等信号通路有关。结论苇茎汤可能通过控制炎症反应、缓解气道高反应及延缓气道重塑等方面治疗哮喘急性发作。
Objective To explore the potential mechanism of Weijing Decoction in the treatment of acute attack of asthma through network pharmacology.Method Use TCMSP database to screen the active components of Weigan decoction.Uniprot and TCMSP database were used to search for the active components and potential targets.Construction of"traditional Chinese medicine-active ingredients-target"network by using Cytoscape.Search the disease target of acute asthma attack in drugbank,genecards,TTD and other databases.The drug activity target of Weijing decoction was intersected with the disease target of acute attack of asthma,and make the Venny map.With the help of STRING database and Cytoscape software,the protein-protein interaction of intersecting targets was analyzed,and PPI map was drawn.The mcode and cytohubba plug-ins of Cytoscape were used to explore the drug disease intersection target relationship.The Metascape analysis platform was used to explore the enrichment of go and KEGG in the intersection targets,and the GO enrichment analysis diagram and KEGG dynamic bubble diagram were drawn using the online mapping platform.Result There were active components in Weijing Decoction and 59 active targets.A total of 827 targets for acute asthma attack were searched,and the intersection of drug disease targets was selected.There were 20 intersection targets,including Jun,CHRM1,CASP8,PTGS2,TGFB1,etc.The results of GO enrichment analysis were:blood circulation,regulation of ion transport,acetylcholine receptor activity,protein domain specific binding and so on.KEGG enrichment analysis showed that Weijing decoction was mainly related to TNF,NF-κB,nerve receptor ligand interaction,PI3 K Akt,calcium ion,nod like receptor signaling pathways.Conclusion The main active components and drug disease intersection targets of Weijing Decoction in the treatment of acute attack of asthma were analyzed by network pharmacology technology,and the biological process and signal pathway of intersection targets were analyzed.It was found that Weijing decoction could control inflammatory reaction,relieve airway hyperresponsiveness and delay airway remodeling to treat acute asthma.
