摘要
为研究藏羊肺脏中缺氧诱导因子1(hypoxia-induciblefactor-1α,HIF-1α)表达特性,揭示藏羊高原适应性,试验选取不同海拔生活的藏羊肺脏组织,采用石蜡切片、HE染色和Masson染色观察肺组织结构,采用免疫组织化学法检测肺脏中HIF-1α的表达。结果表明,较高海拔藏羊肺脏被膜厚度和肺泡平均大小与较低海拔藏羊对比无显著差异(P>0.05),但其被膜中弹性纤维占比、单位面积内肺泡数量、肺泡隔中毛细血管含量、肺泡隔厚度显著高于较低海拔藏羊(P<0.05);HIF-1α在较高海拔藏羊肺脏中,表达强度显著高于较低海拔地区藏羊(P<0.05)。研究表明,藏羊肺脏结构出现适应性改变,且肺脏中HIF-1α蛋白表达与其生存海拔有关。
In order to study the expression of hypoxia inducible factor-1α(HIF-1α)in the Tibetan sheep lung,the lung tissues of Tibetan sheep living at different altitudes were selected,paraffin sections and HE and Masson staining were used to observe the morphological characteristics,immunohistochemistry was used to detect the expression of HIF-1α in the lung.The results showed that there was no significant difference in the thickness of pulmonary capsule and the average size of alveoli between higher and lower altitude living Tibetan sheep(P>0.05).In the higher altitude living Tibetan sheep,the proportion of elastic fiber in lung capsule,the number of alveoli in unit area and the content of capillary in alveolar septum thickness of alveolar septum were significantly higher than those living in lower altitude(P<0.05).The expression of HIF-1α in lungs of higher altitude Tibetan sheep was significantly higher than that of lower altitude ones(P<0.05).The experiment indicates that the lung structure and the expression of HIF-1α in Tibetan sheep was differ when living altitude changes.
