金华猪和长白猪肠道产丁酸菌相对丰度与丁酸代谢的相关性分析 被引量:6

Correlation Analysis between Relative Abundance of Butyrate-Producing Bacteria and Butyrate Metabolism in Intestine of Jinhua and Landrace Pigs

摘要 本试验旨在研究金华猪和长白猪各肠段主要产丁酸菌相对丰度及丁酸代谢的相关性。选取相同条件下饲养的240日龄金华猪和长白猪各10头,分别取不同肠段内容物,采用高通量测序技术分析不同肠段主要产丁酸菌的相对丰度,实时荧光定量PCR法测定产丁酸关键功能基因丁酰-CoA:乙酰-CoA转移酶和丁酸激酶相对表达量的变化,气相色谱法测定丁酸含量。结果表明:1)金华猪空肠和回肠布劳特氏菌属(Blautia)、丁酸球菌属(Butyricicoccus)、丁酸单胞菌属(Butyricimonas)、粪杆菌属(Faecalibacterium)和真杆菌属([Eubacterium]hallii group)相对丰度显著低于长白猪(P<0.05),而颤螺旋菌属(Oscillospira)和罗斯伯里氏菌属(Roseburia)相对丰度显著高于长白猪(P<0.05)。在金华猪和长白猪中,布劳特氏菌属、丁酸球菌属、粪杆菌属和真杆菌属相对丰度间均呈极显著正相关(r=0.48~0.88,P<0.01)。2)金华猪回肠和结肠中丁酰-CoA:乙酰-CoA转移酶和丁酸激酶基因相对表达量显著低于长白猪(P<0.05)。3)金华猪空肠、回肠、结肠中丁酸含量均显著低于长白猪(P<0.05)。4)金华猪和长白猪肠道中丁酰-CoA:乙酰-CoA转移酶和丁酸激酶基因相对表达量与丁酸含量呈极显著正相关(r=0.5704~0.7221,P<0.01);2个品种对比发现,长白猪相关系数均高于金华猪。由此可见,金华猪空肠和回肠产丁酸菌相对丰度显著低于长白猪,同时金华猪结肠产丁酸关键功能基因丁酸激酶和丁酰-CoA:乙酰-CoA转移酶相对表达量显著低于长白猪,与之相对应的是金华猪空肠、回肠和结肠中丁酸含量显著低于长白猪,这也在一定程度上解释了长白猪肠道丁酸含量高于金华猪的原因。 The objective of this study was to investigate the relative abundance of butyrate-producing bacteria and its correlation with butyrate metabolism in the intestine of Jinhua and Landrace pigs.Ten Jinhua pigs and 10 Landrace pigs at 240 days of age raised under the same conditions were selected,and the contents of different intestinal segments were collected respectively.The relative abundance of butyrate-producing bacteria in different intestinal segments were revealed by high throughput sequencing,quantitative PCR was performed to determine the relative expression levels of butyryl-CoA:acetyl-CoA transferase and butyrate kinase genes.The content of butyrate was measured by gas chromatography.The results showed as follows:1)the relative abundance of Blautia,Butyricicoccus,Butyricimonas,Faecalibacterium and[Eubacterium]hallii group in jejunum and ileum of Jinhua pigs were significantly lower than those of Landrace pigs(P<0.05).However,the relative abundance of Oscillospira and Roseburia in Jinhua pigs was significantly higher than that of Landrace pigs(P<0.05).There were significant positive correlations among the relative abundance of Blautia,Butyricicoccus,Faecalibacterium and[Eubacterium]hallii group in Jinhua pigs and Landrace pigs(r=0.48 to 0.88,P<0.01).2)The relative expression levels of butyl-CoA:acetyl-CoA transferase and butyrate kinase genes in the ileum and colon of Jinhua pigs were significantly lower than those of Landrace pigs(P<0.05).3)The contents of butyrate in jejunum,ileum and colon of Jinhua pigs were significantly lower than those of Landrace pigs(P<0.05).4)The relative expression levels of butyl-CoA:acetyl-CoA transferase and butyrate kinase genes were positively correlated with the content of butyrate(r=0.5704 to 0.7221,P<0.01),and the correlation coefficient of Landrace pigs was higher than that of Jinhua pigs.It is concluded that the relative abundance of butyrate-producing bacteria in jejunum and ileum of Jinhua pigs are significantly lower than those of Landrace pigs.Meanwhile,the relative expression levels of butyrate-producing key functional genes of butyrate kinase and butyl-CoA:acetyl-CoA transferase in colon of Jinhua pigs are significantly lower than those of Landrace pigs.Correspondingly,the content of butyrate in jejunum,ileum and colon of Jinhua pigs is significantly lower than that of Landrace pigs.This also explains to some extent that the intestinal butyrate content of Landrace pigs is higher than Jinhua pigs.
