摘要
本试验旨在研究湖羊瘤胃菌群和血浆氧化三甲胺含量对饲粮氯化胆碱含量变化的响应。选取12只体重相近、育肥期健康公湖羊,随机分为对照组和试验组,每组6只。对照组饲喂基础饲粮,试验组饲喂在基础饲粮中添加0.05%氯化胆碱的试验饲粮,试验持续28 d。分别于试验第27和28天晨饲前采集血样和瘤胃内容物。采用靶向代谢组学、高通量测序和实时荧光定量PCR技术对瘤胃菌群和血浆氧化三甲胺含量进行分析。结果显示:与对照组相比,试验组湖羊生长性能和瘤胃发酵参数没有显著差异(P>0.05)。试验组血浆氧化三甲胺含量有升高趋势(P=0.075)。主成分分析(PCoA)结果显示,氯化胆碱改变了细菌菌群结构。线性判别分析(LDA)结果显示,韦荣球菌科(Veillonellaceae)和Oribacterium在对照组中富集,拟杆菌科(Bacteroidaceae)、拟杆菌属(Bacteroides)、柔膜菌纲(Mollicutes)、无壁菌门(Tenericutes)、γ-变形菌纲(Gammaproteobacteria)和柔膜细菌RF9在试验组中富集。实时荧光定量PCR结果显示,氯化胆碱显著促进了瘤胃甲烷马赛球菌目(Methanomassiliicoccales)菌群增殖(P=0.016),但未显著影响胆碱降解细菌cutC基因的丰度(P=0.462)。综上所述,饲粮添加0.05%的氯化胆碱改变了湖羊瘤胃细菌和甲烷菌菌群结构,且有提高血浆中氧化三甲胺含量的趋势。
The aim of this study was to investigate the response of rumen microbiota and plasma trimethylamine N-oxide(TMAO)content to the change of dietary choline chloride content in Hu sheep.Twelve healthy male Hu-sheep with similar body weight in fattening period were randomly arranged into control and experimantal groups,6 in each group.In the control group,sheep were fed a basal diet.In experimental group,sheep were supplemented with 0.05%choline chloride based on the basal diet for 28 days.Blood samples and rumen contents were collected before morning feeding at days 27 and 28,respectively.The results showed as follows:there was no significant difference in growth performance and rumen fermentation parameters between experimental group and control group(P>0.05).The plasma TMAO content and rumen microbial populations were analyzed using targeted metabonomics,high-throughput sequencing and real-time PCR technologies.Results showed that TMAO content in plasma tended to increase in experimental group(P=0.075).PCoA analysis showed that the choline chloride changed the bacterial community structure.LDA analysis showed that Veillonellaceae and Oribacterium enriched in control group,Bacteroidaceae,Bacteroides,Mollicutes,Tenericutes,Gammaproteobacteria and RF9 enriched in experimantal group.Real-time PCR results showed that choline chloride promoted Methanomassiliicoccales proliferation(P=0.016),but did not affect the choline-degrading bacterial cutC gene abundance(P=0.462).The addition of 0.05%choline chloride change the community structure of rumen bacteria and methanogens in Hu sheep,and has a tendency to increase the TMAO content in plasma.
