长期饲喂罗伊氏乳酸杆菌对猪皮下脂肪及肝脏脂肪代谢相关基因表达的影响

Effects of Long-Term Feeding of Lactobacillus reuteri on expression of Genes Related to Lipid Metabolism in Subcutaneous Fat and Liver of Pigs

摘要 本试验旨在研究长期饲喂罗伊氏乳酸杆菌(Lactobacillus reuteri 1,LR1)对猪皮下脂肪及肝脏脂肪代谢相关基因表达的影响。试验选用144头21日龄三元(杜×长×大)杂交断奶仔猪,随机分为3组,每组8个重复,每个重复6头猪。对照1组饲粮基础饲粮,对照2组饲喂在基础饲粮中添加抗生素的试验饲粮,LR1组饲喂在基础饲粮中添加5×10^(10) CFU/kg LR1的试验饲粮,试验期175 d。结果表明:1)与对照1组相比,对照2组的腹脂胆固醇调节元件结合蛋白(SREBP)、乙酰辅酶A羧化酶(ACC)、脂肪酸合成酶(FAS)、硬脂酰辅酶A去饱和酶(SCD)及甘油三酯脂酶(ATGL)基因相对表达量显著增加(P<0.05),肉毒碱棕榈酰转移酶-1β(CPT-1β)、过氧化物酶体增殖物激活受体α(PPARα)、脂肪酸转运蛋白-1(FATP-1)及过氧化物酶体增殖物激活受体γ(PPARγ)基因相对表达量无显著差异(P>0.05);LR1组腹脂FATP-1基因相对表达量显著增加(P<0.05),SREBP、ACC、CPT-1β、FAS、PPARα、SCD、PPARγ及ATGL基因相对表达量无显著差异(P>0.05);与对照2组相比,LR1组SREBP、ACC及SCD基因相对表达量显著降低(P<0.05)。2)与对照1组相比,对照2组背脂SCD及ATGL基因相对表达量显著增加(P<0.05),而LR1组背脂SCD基因相对表达量显著降低(P<0.05);与对照2组相比,LR1组背脂ACC、FAS及SCD基因相对表达量显著降低(P<0.05);与对照1、2组相比,LR1组背脂PPARγ基因相对表达量显著提高(P<0.05)。3)与对照1组相比,对照2组及LR1组肝脏SREBP、ACC、FAS及SCD基因相对表达量均显著降低(P<0.05),而CPT-1β、PPARα、FATP-1、PPARγ及ATGL基因相对表达无显著变化(P>0.05)。综上所述,LR1促进猪皮下脂肪的脂肪酸转运,较抗生素显著降低了猪背脂及肝脏脂肪合成代谢。此外,LR1不影响肝脏的转运及分解代谢。 This experiment was aimed to study effects of Lactobacillus reuteri 1(LR1)on expression of genes related to lipid metabolism in subcutaneous fat and liver of pigs.A total of 144 cross-bred(Duroc×Landrace×Yorkshire)21-day-old weaned piglets were randomly assigned to 3 groups,each group comprised 8 replicate pens with 6 pigs per pen.The diet in the control 1 group was a basal diet,the diet in the control 2 group was the basal diet supplemented with antibiotics,and the diet in LR1 group was the basal diet supplemented with 5×10^(10) CFU/kg LR1.The experiment period was 175 days.The results showed as follows:1)compared with the control 1 group,the relative expression levels of sterol regulatory element binding protein(SREBP),acetyl CoA carboxylase(ACC),fatty acid synthase(FAS),stearoyl CoA desaturase(SCD)and adipose triglyceride lipase(ATGL)genes in abdominal adipose in the control 2 group were significantly increased(P<0.05),but there were no significant difference in the relative expression levels of carnitine palmitoyl transferase 1 beta(CPT-1β),peroxisome proliferator-activated receptor alfa(PPARα),fatty acid transport protein-1(FATP-1),peroxisome proliferator-activated receptor gamma(PPARγ)genes(P>0.05);the relative expression level of FATP-1 gene was increased significantly in LR1 group(P<0.05),and the relative expression levels of SREBP,ACC,CPT-1β,FAS,PPARα,SCD,PPARγand ATGL genes had no significant different(P>0.05);the relative expression levels of SREBP,ACC and SCD genes in LR1 group were significantly decreased compared with the control 2 group(P>0.05).2)Compared with control 1 group,the relative expression levels of SCD and ATGL genes in back fat was increased significantly in the control 2 group(P<0.05),and the relative expression level of SCD gene was decreased significantly in LR1 group(P<0.05);compared with control 2 group,the relative expression levels of ACC,FAS and SCD genes in the LR1 group were significantly reduced(P<0.05);compared with the control 1 and 2 groups,the relative expression level of PPARγgene in backfat in LR1 group was significantly increased(P<0.05).3)Compared with control 1 group,the relative expression levels of SREBP,ACC,FAS and SCD genes in liver in the control 2 group and LR1 group were significantly decreased(P<0.05),but the relative expression levels of CPT-1β,PPARα,FATP-1,PPARγand ATGL genes had no significant differences(P>0.05).In conclusion,LR1 promotes fatty acids transport in subcutaneous fat and decreases anabolism of backfat and liver in pigs compared with antibiotics.Additionally,LR1 affects lipid transport and catabolism of liver in pigs.
