摘要
[目的]研究含多种中药成分的抗菌喷雾的体外抗菌效果并对其抗菌机理进行分析。[方法]选用大肠杆菌(E.coli)和金黄色葡萄球菌(S.aureus)2种菌,用肉汤稀释法和荧光染色法测定抗菌喷雾对其抑菌活性、抑菌率、最小抑菌浓度(MIC)和最低杀菌浓度(MBC),用滤纸片法测定其抑菌环,用扫描电子显微镜(SEM)和透射电子显微镜(TEM)观察抗菌喷雾对细菌表面结构和超微结构的影响。[结果]中药抗菌喷雾在极短的时间内对E.coli和S.aureus的抑菌率就能达到100%,滤纸法显示抑菌环明显;MIC和MBC结果说明了中药抗菌喷雾在稀释5倍之后仍对2种细菌具有抑制作用;喷雾作用后的细菌大量死亡,表面及内部结构均发生明显变化。[结论]多种中药成分的抗菌喷雾对E.coli和S.aureus具有显著的联合抑菌效果。
[Objective]To study the in vitro antibacterial effect of an antibacterial spray which contains several kinds of traditional Chinese medicine and analyze its antibacterial mechanism.[Method]Escherichia coli(E.coli)and Staphylococcus aureus(S.aureus)were selected to determine the antibacterial activity,antibacterial rate,minimum inhibitory concentration(MIC)and minimum bactericidal concentration(MBC)of antibacterial spray by broth microdilution method and fluorescence staining method.The inhibition zone was determined by filter paper method.The effects of antibacterial spray on the surface structure and ultrastructure of bacteria were observed by scanning electron microscope(SEM)and transmission electron microscope(TEM).[Result]The antibacterial rate of traditional Chinese medicine antibacterial spray against E.coli and S.aureus could reach 100%in a very short time.Filter paper method demonstrated obvious inhibition zone.The results of MIC and MBC indicated that the traditional Chinese medicine antibacterial spray against E.coli and S.aureus still had two kinds of bacteria inhibition effect after 5 times dilution.After spraying,a large number of bacteria died,and the surface and internal structure changed evidently.[Conclusion]The antibacterial spray of various kinds of Chinese medicine has remarkable bacteriostatic effect on E.coli and S.aureus.
