摘要
为了筛选高效利用甲醇的功能性微生物,从气化厂甲醇车间附近的土壤中分离筛选菌株,结合形态特征、生理生化特性和16S rDNA序列对其进行鉴定,并对菌株生长及甲醇降解条件进行研究。结果表明,分离得到一株细菌,编号为4311,经鉴定为甲基球形菌属(Methylopila sp.)。该菌株最适生长条件为初始甲醇体积分数1.0%,初始pH值5.0,氯化钠添加量0.05%,氮源为蛋白胨,其添加量为2 g/L。在此优化条件下,菌株4311对1.0%(V/V)甲醇的降解率达到85%。
In order to screen the functional microorganisms that can efficiently utilize methanol,a strain of bacterium numbered 4311,was isolated and screened from the soil near the methanol workshop of a gasification plant.According to the results of morphological,biochemical and 16S rDNA sequence analysis,strain 4311 was identified as Methylopila sp.The optimal growth conditions of strain 4311 were initial methanol concentration 1.0%(V/V),initial pH 5.0,sodium chloride concentration 0.05%.The peptone was the optimal nitrogen source,and optimal peptone dosage was 2 g/L.Under these optimized conditions,the degradation rate of 1.0%(V/V)methanol by strain 4311 reached 85%.
