摘要
以香菇柄为主要原料,采用纤维素酶和糖化酶双酶同步酶解工艺,通过单因素试验和Box-Behnken响应面试验优化香菇柄糖化工艺。结果表明,双酶同步糖化最佳工艺条件为:纤维素酶和糖化酶酶活比5∶3、双酶添加量3164 U/g、料液比1∶15(g∶mL)、酶解时间120 min,酶解温度50℃。在此优化条件下,香菇柄水解液还原糖得率达11.05%。香菇柄双酶同步糖化条件温和、安全性高,可提高香菇柄还原糖含量,可为香菇柄无添加糖酿造酒等发酵食品的开发提供参考。
Using Lentinus edodes stem as main raw material,the synchronously bienzymatic saccharification technology with cellulose and glucoamylase was optimized by single factor and Box-Behnken response surface experiments.The results showed that the optimum synchronous bienzymatic saccharification conditions were obtained as follows:cellulose and glucoamylase activity ratio 5:3,compound enzymes addition 3164 U/g,material and liquid ratio 1∶15(g∶ml),enzymolysis time 120 min,and temperature 50℃.Under these optimal conditions,the yield of reducing sugar from L.edodes stem hydrolysate reached 11.05%.The synchronously bienzymatic saccharification conditions of L.edodes stem were mild,highly safe,and could increase the reducing sugar content of L.edodes stem,which could provide a reference for the development of fermented foods such as sugar-free L.edodes stem alcoholic drink.
