CD19^(+)CD24^(high)CD27^(+)调节性B细胞在强直性脊柱炎检测中的临床意义

摘要 目的拟对AS患者外周血中的调节性B细胞进行检测,并对其临床意义进行分析。探讨B细胞是否具有免疫抑制功能及其发生机制。方法采集22例HLA-B-27阳性AS患者和22例非AS健康对照者的外周血标本,用CD3、CD4、CD19、CD24和CD27抗体对外周血进行染色,红细胞溶解液进行红细胞破碎,并用流式细胞仪对CD19^(+)CD24^(high)CD27^(+)调节性B细胞检测,对AS患者外周血清进行IL-10浓度检测。与AS患者血沉和疾病活动度作相关性分析。结果与22例非AS对照组相比,22例AS患者CD19^(+)CD24^(high)CD27^(+)调节性B细胞在AS患者中显著上升,与血沉、疾病活动度及外周血清IL-10浓度正相关。结论 AS患者外周血CD19^(+)CD24^(high)CD27^(+)调节性B细胞显著上升,且上升频率与疾病活动度显著相关。 Objective To discuss the immunosuppressive function of B cell and its mechanism.Present study is mainly focused on tumor patients,while there are few studies on the chronic inflammatory rheumatic disease of ankylosing spondylitis(AS).Methods Peripheral blood samples were collected from 22 HLA-B-27 positive AS patients and 22 non-AS healthy controls,and stained with CD3,CD4,CD19,CD24 and CD27 antibodies,then the red blood cells were crushed with erythrocyte lysate,the CD19^(+)CD24^(high)CD27^(+)regulatory B cells were detected by the flow cytometer.The concentration of IL-10 in peripheral blood of AS patients was detected.The correlation between erythrocyte sedimentation rate and disease activity in AS patients was analyzed.Results Compared with 22 non-AS controls,CD19+CD24highCD27+regulatory B cells were significantly increased in 22 AS patients,which was positively correlated with erythrocytic sedimentation rate,disease activity and IL-10 concentration in peripheral serum.Conclusion CD19^(+)CD24^(high)CD27^(+)regulatory B cells was significantly increased in AS patients,which also positively associated with the disease activity.
