芪蛭胶囊对大鼠脑缺血再灌注损伤相关因子p-Akt、Cathepsin表达的影响 被引量:1

Effects of Qizhi Capsules on the Expression of p-Akt,Cathepsin Related Factors in Rats with Cerebral Ischemia Reperfusion Injury

摘要 目的通过对PI3K-Akt-mTOR信号通路相关因子p-Akt、Cathepsin B、Cathepsin D表达的研究,进一步探讨芪蛭胶囊对大鼠脑缺血再灌注损伤神经功能的保护作用。方法将60只雄性SD大鼠随机分为假手术组、模型组、3-MA组、LY组和中药组。制备SD大鼠大脑中动脉阻塞(MCAO)模型,缺血2 h后进行再灌注。各组给予相应药物干预,连续7 d,第8日造模。应用神经功能评分法(NSS)评估神经功能缺损程度,脑组织含水量检测脑水肿情况,Western blotting检测相关因子p-Akt、Cathepsin B、Cathepsin D表达水平。结果同模型组比较,中药组神经功能评分明显降低(P<0.05),中药组脑组织含水量降低(P<0.05);Western blotting结果显示:与模型组相比,中药组中p-Akt表达减少(P<0.05),中药组Cathepsin B、Cathepsin D表达显著增加(P<0.05)。结论芪蛭胶囊可下调脑缺血再灌注损伤大鼠神经功能评分、脑组织含水量,可降低p-Akt的表达,增加Cathepsin B、Cathepsin D的表达,进一步激活自噬,进而保护脑缺血再灌注损伤后大鼠脑神经功能。 Objective:To observe the protective effect of Qizhi Capsules on the neurological function of rats with cerebral ischemia reperfusion injury through the autophagy-related factors p-AKT,Cathepsin B,Cathepsin D of PI3 K-Akt-mTOR signaling pathway.Methods:A total of 60 SD male rats were randomly divided into sham group,model group,3-MA group,LY group and TCM group.The middle cerebral artery occlusion model(MCAO)was made in the SD rats,and reperfusion was performed 2 hours after ischemia.Each group was administered corresponding drug intervention for 7 days,and the model was made on the 8 th day.The degree of nerve functional defects was detected by neurological severity score(NSS),the condition of cerebral edema was detected the water content of brain tissue,and the expression of autophagy-related factors p-Akt,Cathepsin B,Cathepsin D was detected by Western blotting.Results:The neurologic severity score was significantly lower in the TCM group than that in the model group(P<0.05);the water content of brain tissue was lower in the TCM group than that in the model group(P<0.05);Compared to the model group.Western blotting showed that the expressions of p-Akt were decreased in the TCM group(P<0.05),while Cathepsin B,Cathepsin D were increased in the TCM group(P<0.05).Conclusion:Qizhi Capsules can decrease neurological severity score and the water content of brain tissue of cerebral ischemia reperfusion injury rats.It can also decrease the expression of p-Akt,increase the expression of Cathepsin B,Cathepsin D,and further activate autophagy and then protect the function of brain nerve in rats with cerebral ischemia reperfusion injury.
