摘要
目的探究白介素-1(IL-1)、Toll样受体4(TLR4)、E-钙黏蛋白(E-cadherin)基因在巨细胞病毒感染性流产中的表达及意义。方法选取2019年1月-2020年1月九江市妇幼保健院产科收治的流产患者263例作为流产组,根据患者是否为巨细胞病毒感染所致的流产分为感染流产组(125例,巨细胞病毒感染性流产)和非感染流产组(138例,要求人工流产终止妊娠的正常单胎自然受孕产妇),同期选取于医院行健康体检的正常妊娠产妇80名作为正常妊娠组。采用酶联免疫吸附试验法测定所有研究对象血清IL-1表达水平,采用实时荧光定量PCR(RT-PCR)方法检测所有研究对象外周血单个核细胞TLR4 mRNA、E-cadherin mRNA表达量,分析IL-1、TLR4、E-cadherin基因在巨细胞病毒感染性流产中的表达及意义。结果与正常妊娠组相比,流产组产妇血IL-1表达水平、TLR4 mRNA、E-cadherin mRNA表达量分别为(25.59±1.41)ng/L、(1.22±0.19)、(1.40±0.13)均升高,具有统计学差异(P均<0.05)。与非感染流产组相比,感染流产组产妇血IL-1表达水平、TLR4 mRNA、E-cadherin mRNA表达量分别为(37.08±2.54)ng/L、(1.71±0.30)、(1.93±0.40)均升高,具有统计学差异(P均<0.05)。与非活动性感染相比,活动性感染产妇血IL-1表达水平、TLR4 mRNA、E-cadherin mRNA表达量分别为(48.24±4.05)ng/L、(2.03±0.29)、(2.54±0.14)均升高,具有统计学差异(P均<0.05)。结论 IL-1、TLR4、E-cadherin基因在巨细胞病毒感染性流产中呈现为高表达,且活动性感染表达更为加剧,与巨细胞病毒感染的病理过程密切相关,可作为产妇巨细胞病毒感染的检测指标。
OBJECTIVE To explore the expression and significance of IL-1, TLR4 and E-cadherin genes in cytomegalovirus induced abortion. METHODS Total of 263 cases of abortion patients admitted to Jiujiang maternal and child health hospital from Jan. 2019 to Jan. 2020 were enrolled as the abortion group. According to whether cytomegalovirus infection was detected in the patients, they were divided into two subgroups, the infected abortion group(125 cases with cytomegalovirus infected abortion) and non-infected abortion group(138 cases of normal singleton spontaneous pregnant women requiring abortion termination of pregnancy), and another 80 normal pregnant women under physical examination in our hospital were taken as the normal pregnancy group. Enzyme linked immunosorbent assay(ELISA) was used to detect protein expression of IL-1 in all subjects. Real time PCR was used to detect the mRNA expression of TLR4 and E-cadherin in peripheral blood mononuclear cells of all subjects. The expression significance of IL-1, TLR4 and E-cadherin genes in cytomegalovirus induced abortion were analyzed. RESULTS The blood IL-1 level, TLR4 and E-cadherin mRNA expression levels in abortion group were(25.59±1.41)ng/L,(1.22±0.19) and(1.40±0.13), respectively, which were significantly higher than those in the normal pregnancy group(all P<0.05). The blood IL-1 expression level and TLR4 and E-cadherin mRNA expression levels in the infected abortion group were(37.08±2.54)ng/L,(1.71±0.30) and(1.93±0.40), respectively, which were significantly higher than those in the non-infected abortion group(all P<0.05). The blood IL-1 expression level and TLR4 and E-cadherin mRNA expression levels in active infected parturient were(48.24±4.05)ng/L,(2.03±0.29) and(2.54±0.14), respectively;the differences among them were significant(all P<0.05). CONCLUSION Expression levels of IL-1, TLR4 and E-cadherin are high in women with cytomegalovirus infected abortion, especially in the active infection patients, which is closely related to the pathological process of cytomegalovirus infection and can be recommended as the detection indexes of maternal cytomegalovirus infection.
