基于NO_(3)^(-)-NO_(2)^(-)-NO通道探索白酒在瓜蒌薤白白酒汤保护小鼠缺血心肌中的佐使作用 被引量:4

摘要 目的通过检测瓜蒌薤白白酒汤各组分中药及其配伍药对小鼠缺血心肌组织病理学及血清一氧化氮(NO)含量的影响,初步探索瓜蒌薤白白酒汤对小鼠缺血心肌的保护作用及白酒的佐使作用内涵。方法采用HE染色法检测小鼠缺血心肌的组织病理学变化;利用Griess法检测各组血清中NO的含量。结果与心肌梗死模型组相比,瓜蒌、薤白和白酒组可见部分心肌细胞较完整,部分心肌纤维有断裂,部分胞核散落在细胞外;当瓜蒌或薤白与白酒配伍后,可见大部分心肌细胞较完整,轮廓清晰,大部分胞核形态正常;瓜蒌薤白白酒汤组心肌细胞基本完整,轮廓清晰,仅可见个别心肌细胞边缘不清;与心肌梗死模型组相比,配伍组血清中的NO水平明显升高(P<0.05),白酒组差异无统计学意义(P>0.05);与瓜蒌组和薤白组相比,配伍组血清中的NO含量明显升高(P<0.05),瓜蒌薤白白酒汤组的NO含量最高,与所有其他各组相比均差异有统计学意义(P<0.01)。结论该研究证实了瓜蒌、薤白和白酒对小鼠缺血心肌具有不同程度的保护作用,当白酒与富含硝酸盐/亚硝酸盐(NO_(3)^(-)/NO_(2)^(-))的瓜蒌、薤白配伍后,通过NO_(3)^(-)-NO_(2)^(-)-NO通道产生更为强烈的NO生物活性,保护小鼠缺血心肌,在瓜蒌薤白白酒汤中发挥了显著的佐使作用。 Objective To investigate the effects of different components of Gualou Xiebai Baijiu Decoction(瓜蒌薤白白酒汤)and their compatibility on the histopathology of ischemic myocardium and the content of serum nitric oxide(NO)in mice,and explore the protective effect of Gualou Xiebai Baijiu Decoction on ischemic myocardium in mice and the adjuvant effect of white liquor.Methods Hematoxylineosin(HE)staining was used to detect the histopathological changes of ischemic myocardium in mice;The content of NO was quantified by Griess method.Results Compared with the myocardial infarction model group,Gualou(Trichosanthis Fructus,GL),Xiebai(Allii Macrostemonis Bulbus,XB),and white liquor(WL)groups showed that some myocardial cells were relatively intact,some myocardial fibers were broken,some nuclei were scattered outside the cells.Once white liquor combined with Gualou(Trichosanthis Fructus)and Xiebai(Allii Macrostemonis Bulbus),or both of them,cardiomyocytes were relatively complete,with clear contours and most of the nuclei were normal in shape.GLXBBJD group is basically complete with clear contours and only a few myocardial cell edges are unclear.Compared with myocardial infarction model group,NO level of GL group and XB group was significantly increased(P<0.05),no difference in WL group(P>0.05).Compared with GL group and XB group,NO content in serum of compatibility group was significantly increased(P<0.05).The GLXBBJD group had the highest NO content,which was significantly different from all other groups(P<0.01).Conclusion This study confirms that Gualou(Trichosanthis Fructus),Xiebai(Allii Macrostemonis Bulbus)and white liquor have different degrees of protective effects on ischemic myocardium in mice.The NO_(3)^(-)-NO_(2)^(-)-NO pathway produces stronger NO biological activity to protect the ischemic myocardium in mice once white liquor combined with Gualou(Trichosanthis Fructus)and Xiebai(Allii Macrostemonis Bulbus)or both of them.White Liquor significantly plays a adjuvant role in GLXBBJD.
