基于网络药理学探讨黄芪-黄连药对治疗冠心病作用机制 被引量:3

Mechanism Research of Huangqi(Astragali Radix)-Huanglian(Coptidis Rhizoma) in Treatment of Coronary Heart Disease Based on Network Pharmacology

摘要 目的通过网络药理学方法探讨黄芪-黄连药对治疗冠心病的作用机制。方法利用中药系统药理学数据库与分析平台(TCMSP)检索黄芪-黄连药对的化学成分和作用靶点,通过Cytoscape 3.7.1绘制活性成分-靶点网络图。利用DisGeNET、GeneCards数据库检索冠心病相关靶点,取交集靶点上传至STRING11.0数据库构建蛋白互作(PPI)网络,进一步筛选关键靶点。利用Metascape平台对关键靶点进行基因本体论(gene ontology,GO)和京都基因百科全书(kyoto encyclopedia of genes and genomes,KEGG)功能富集分析。结果共获取黄芪-黄连药对活性成分37个,对应靶点213个。冠心病靶点1063个,药物疾病作用关键靶点30个,包括AKT1、IL-6、TNF、VEGFA、TP53等,KEGG分析显示其作用潜在通路包括AGE-RAGE信号通路、流体剪切应力与动脉粥样硬化、IL-17信号通路等。结论本研究初步揭示了黄芪-黄连药对治疗冠心病的多成分、多靶点、多通路作用机制,为进一步探究益气解毒法治疗冠心病提供理论基础与研究思路。 Objective To explore the mechanism of the treatment of coronary heart disease with Huangqi(Astragali Radix)-Huanglian(Coptidis Rhizoma)via network pharmacology. Methods TCMSP platform was used to retrieve the chemical components and targets of Huangqi(Astragali Radix)and Huanglian(Coptidis Rhizoma),the active ingredient-target network was drawn through Cytoscape 3.7.1. Using the DisGeNET and GeneCards databases to search for coronary heart disease-related targets,take the intersection targets and upload them to the STRING11.0 database to construct a protein interaction(PPI)network to further screen key targets. Using Metascape platform to carry out GO function enrichment analysis and KEGG pathway enrichment analysis on key targets. Results A total of 37 active components of Huangqi(Astragali Radix)-Huanglian(Coptidis Rhizoma)were obtained,corresponding to 213 targets. There were 1063 targets for coronary heart disease and 30 key targets for drug disease,including AKT1,IL-6,TNF,VEGFA,TP53,etc. KEGG analysis showed that its potential pathways include AGE-RAGE signaling pathway,fluid shear stress and atherosclerosis,IL-17 signaling pathway,etc. Conclusion This study has initially revealed the multi-component,multi-target,and multi-path mechanism of Huangqi(Astragali Radix)-Huanglian(Coptidis Rhizoma)medicine in the treatment of coronary heart disease,which provides a theoretical basis and research ideas for further exploration benefitting Qi and removing virus method in the treatment of coronary heart disease.
