头穴丛刺法对局灶性脑缺血大鼠突触形态可塑性的影响 被引量:8

摘要 目的探讨头穴丛刺法对MCAO大鼠的突触可塑性的影响,为临床应用头穴丛刺法治疗脑卒中提供理论依据。方法将27只SD大鼠随机分为头穴丛刺、模型组与假手术组;将头穴丛刺组与模型组的大鼠制备大脑中动脉缺血模型。于造模当天起,头穴丛刺组予每日一次的头穴丛刺治疗,模型组与假手术组不做治疗。分别在造模后1 d、7 d、14 d进行大鼠神经功能评分并取材。透射电镜观察大脑皮层半暗带区突触间隙宽度、突触界面曲率及突触后致密物厚度的变化。结果 (1)各时间点头穴丛刺组神经功能评分均优于模型组,但1 d时,头穴丛刺组、模型组组间比较无统计学意义(P>0.05);7 d、14 d时,具有统计学意义(P<0.05)。(2)1 d时:头穴丛刺组、模型组的突触各形态学参数与假手术组相比,均无明显差异(P>0.05);7 d时:头穴丛刺组突触后致密物厚度明显高于模型组,具有统计学意义(P<0.05);14 d时,头穴丛刺组的突触后致密物厚度、突触间隙宽度与模型组相比,具有统计学意义(P<0.05)。各组大鼠各时间点突触界面曲率比较均无统计学意义(P>0.05)。结论头穴丛刺法具有一定程度的促进突触形态可塑性的作用。 Objective To study the effect of cluster needling of scalp acupuncture on middle cerebral artery occlusion(MCAO) rats′ synaptic plasticity.To search the theoretical basis of clinical cluster needling therapy for ischemic stroke.Methods A total of 27 SD rats were randomly divided into cluster needling group,model group,and sham operation group.The cluster needling group and model group were made to be middle cerebral artery occlusion model.From the day of establishment,the cluster needling group received treatment once per day while the model group and sham operation group received no therapy.All groups received neurological function scoring and were drawn materials at 1st,7th,14th day after establishment.The observation of the cerebral cortex penumbra by transmission electron microscope included synaptic plasticity,the width of synapsis cleftand the thickness of post synaptic density was carried out.Results(1)Neurological function score:The scores of the cluster needling group on the 7th and 14th day were both obviously higher than the model group′s(P<0.05).(2)Morphological parameters:On the 7st day,the thickness of post synaptic density of cluster needling group was better than the model group′s(P<0.05).On the 14th day,both the thickness of post synaptic density and the width of synapsis cleft of the cluster needling group were better than the model group′s(P<0.05).The comparisons of the curvature of synaptic interface among these groups all had no significant difference(P>0.05).Conclusion The cluster needling of scalp acupuncture therapy can improve synaptic plasticity in certain extent.
