尖叶假龙胆对脑缺血再灌注损伤大鼠保护作用研究 被引量:1

Research of Protective Effect of Jianyejialongdan(Gentianella Acuta)Acuminata on Cerebral Ischemia Reperfusion Injury in Rats

摘要 目的研究不同组分尖叶假龙胆对脑缺血再灌注损伤大鼠的保护作用。方法将90只健康雄性大鼠分为9组,分别为空白组、模型组、假手术组、阳性药组、全草组、水洗脱组、30%乙醇洗脱组、60%乙醇洗脱组、90%乙醇洗脱组。其中空白组、模型组、假手术组给予等量生理盐水4周,其余组给予相应药物灌胃4周。第5周开始运用线栓法制备大鼠脑缺血再灌注损伤模型,在大鼠缺血2 h再灌注22 h的情况下检测各组大鼠脑组织中的丙二醛(MDA)、超氧化物歧化酶(SOD)的含量,凋亡相关蛋白活性,及血清中炎性因子的表达水平,并通过HE染色观察大鼠脑细胞的病理变化。结果在大鼠发生脑缺血再灌注损伤后,其脑组织及生化指标出现异常,HE染色显示给药组的大鼠梗死侧大脑皮质出现经典的梗死样改变,但与模型组对比,给药组的细胞损伤较轻,不同浓度尖叶假龙胆的提取组分均能减轻脑缺血再灌注损伤大鼠的病变反应,促使脑组织中MDA浓度降低,SOD活力提升,Bcl-2蛋白表达提高,Bax和Caspase-3蛋白表达降低,血清中炎症因子白细胞介素1(IL-1)、白细胞介素6(IL-6)含量降低,其中90%乙醇洗脱组疗效更佳。结论尖叶假龙胆对脑缺血再灌注损伤大鼠具有保护作用,其部分机制可能与抗氧化应激、减轻细胞凋亡及抑制炎症反应有关。 Objective To study the protective effects of different components of Jianyejialongdan(Gentianella Acuta)on rats with cerebral ischemia-reperfusion injury. Methods 90 healthy male rats were divided into nine groups:blank group,model group,sham operation group,positive drug group,whole grass group,water washing group,30% ethanol elution group,60% ethanol elution group and 90% ethanol elution group. The blank group,model group and sham operation group were given the same amount of saline for 4 weeks,and the other groups were given the corresponding drugs for 4 weeks. In the fifth week,the rat model of cerebral ischemia-reperfusion injury was established by using the method of thread bolt. The content of MDA and SOD,the activity of apoptosis related protein and the expression of inflammatory factors in serum were measured in each group after 2 hours of ischemia-reperfusion for 22 hours,and the pathological changes of brain cells were observed by HE staining. Results After the cerebral ischemia-reperfusion injury,the brain tissue and biochemical indexes of the rats were abnormal. HE staining showed that the cerebral cortex in the infarcted side of the rats in the administration group had classic infarct like changes,but compared with the model group,the cells in the administration group were slightly damaged. The extracts of Jianyejialongdan(Gentianella Acuta)at different concentrations could reduce the pathological reaction of the rats with cerebral ischemia-reperfusion injury and promote the pathological reaction. The concentration of MDA in brain decreased,the activity of SOD increased,the expression of Bcl-2 protein increased,the expression of Bax and Caspase-3 protein decreased,the content of IL-1 and IL-6 in serum decreased,and 90% ethanol elution group had better effect. Conclusion Jianyejialongdan(Gentianella Acuta)has protective effect on rats with cerebral ischemia-reperfusion injury,and part of its mechanism may be related to anti oxidative stress,reducing apoptosis and inhibiting inflammatory response.
