不同生长年限霍山石斛抗炎和抗肿瘤作用比较研究 被引量:4

摘要 目的:研究不同生长年限霍山石斛活性成分多糖、黄酮、生物碱和石斛酚的含量差异以及霍山石斛抗炎、对HeLa细胞增殖的抑制作用。方法:以二甲苯建立小鼠耳肿胀模型,C57BL/6小鼠随机分为空白组、地塞米松组、不同生长年限霍山石斛榨汁液高、中、低剂量组,每日灌胃给药,连续一周,计算肿胀度和肿胀抑制率。HeLa细胞随机分为不同生长年限不同浓度的霍山石斛水提物组、阳性对照组、空白组,CCK-8法检测各细胞增殖情况。结果:不同生长年限的霍山石斛都能够降低二甲苯所致的耳肿胀度,二年生高剂量霍山石斛组抑制效果最好。不同生长年限霍山石斛水提物作用于HeLa细胞24 h后,质量浓度在2~10 mg/mL范围内均可抑制HeLa细胞活力,且随着浓度的升高,细胞增殖抑制作用增强。结论:2年生霍山石斛抗炎效果最佳,3年生霍山石斛抗肿瘤效果最好。 Objective:To study the content differences of polysaccharides,flavonoids,alkaloids and dendrobiol in different growth years of Dendrobium huoshanense,as well as the anti-inflammatory and inhibition of HeLa cell proliferation of D.huoshanense with different growth years.Methods:A mouse ear swelling model was established with xylene.C57BL/6 mice were randomly divided into a blank group,a dexamethasone group,and high,medium,and low dose groups of D.huoshanense juice in different growth years.They were given daily by intragastric administration for one week and the swelling degree and swelling inhibition rate were calculated.HeLa cells were randomly divided into different growth years and different concentrations of D.huoshanense water extract group,positive control group and blank group.CCK-8 method was used to detect cell proliferation.Results:D.huoshanense with different growth years could reduce the ear swelling induced by xylene,and the ear swelling inhibition effect of high-dose biennial D.huoshanense was the best.After different growth years of D.huoshanense were treated on HeLa cells for 24 h,the mass concentration of 2-10 mg/mL could inhibit the viability of HeLa cells,and with the increase of the concentration,the cell proliferation inhibition increased.Conclusion:Biennial D.huoshanense has the best anti-inflammatory effect,and triennial D.huoshanense has the best anti-tumor effect.
