摘要
用从鲍姆纤孔菌桑黄(Sanghuangporus baumii)液体发酵菌丝体中获得的菌丝三萜(SGC),研究其对小鼠腹腔巨噬细胞RAW264.7免疫调节活性的影响。利用体外建立巨噬细胞RAW264.7炎症模型,用不同浓度的菌丝三萜处理细胞,采用Cell Counting Kit 8(CCK-8)和酶联免疫方法测定细胞增殖、细胞活力和细胞因子的分泌变化。结果显示:菌丝三萜对巨噬细胞RAW264.7的生长没有毒性,细胞活力试验显示菌丝三萜可提高溶菌酶、谷胱甘肽和糖原的含量;使得ATP酶含量降低;高浓度的菌丝三萜可以促进巨噬细胞分泌一氧化氮(NO);能够抑制脂多糖(LPS)诱导下小鼠巨噬细胞分泌白细胞介素IL-1α、IL-6、肿瘤坏死因子TNF-α和INF-r的含量。说明在巨噬细胞受到外界刺激后,SGC对其具有一定的保护作用。
Obtain mycelium triterpenes by solid fermentation of Sanghuangporus baumii,and then its effect on immunomodulatory activity of mouse peritoneal macrophage RAW264.7 was studied.The macrophage RAW264.7 inflammation model was established in vitro,cells were treated with different concentrations of mycelium triterpenes extractions,and cell proliferation,cell viability and cytokine secretion changes were determined by CCK-8 and enzyme-linked immunoassay.The result show that mycelium triterpenes extractions is not toxic to the growth of macrophage RAW264.7.Cell viability experiments showed that mycelium triterpenes extractions can increase the content of lysozyme,glutathione and glycogen,and reduced the content of acidic ATPase.High concentration of mycelium triterpenes extractions can promote NO secretion by macrophages;and that can inhibit the levels of IL-1α,IL-6,TNF-αand INF-r secreted by mouse macrophages induced by lipopolysacch aride.In the correlation analysis,lysozyme,glutathione and glycogen were highly positively correlated;IL-1αwas highly positively correlated with IL-6 and INF-r.Indicate that Triterpenoids from Sanghuangporus baumii plays a certain role in protecting the body from external stimulation.
