痔疾洗液联合红光治疗对大鼠肛周创面模型术后恢复的影响 被引量:2

Effect of Hemorrhoid Lotion combined with red light therapy on postoperative recovery of perianal wound model in rats

摘要 目的观察痔疾洗液联合红光治疗大鼠肛周创面模型的疗效及其病理机制。方法大鼠按随机数字表法分为空白对照组、模型对照组、痔疾洗液组、红光治疗组和痔疾洗液+红光治疗组。制备大鼠肛周创面模型,各组分别予以对应治疗。2周后观察大鼠一般情况;测定各组创面愈合率以及创面上皮化的动物数量;使用实时荧光定量核酸扩增检测系统(qPCR)检测大鼠血清IL-8、TNFα、TGF-β1、EGF的表达及变化;酶联免疫吸附法(ELISA)检测大鼠皮肤组织IL-8、TNFα、TGF-β1、EGF表达及变化。结果与模型对照组比较,痔疾洗液组和红光治疗组创面愈合率明显改善(P<0.05),创面上皮化动物数量明显增多(P<0.05);痔疾洗液+红光治疗组创面愈合率则进一步改善(P<0.01),创面上皮化的动物数量进一步增多(P<0.01)。模型对照组中的IL-8、TNFα的表达显著上调(P<0.01),TGF-β1、EGF/p-EGFR的表达显著下调(P<0.01);与模型对照组比较痔疾洗液组和红光治疗组的IL-8、TNFα的表达显著下调(P<0.05),TGF-β1、EGF/p-EGFR的表达显著上调(P<0.05);痔疾洗液+红光治疗组的IL-8、TNFα、TGF-β1、EGF/p-EGFR趋势则进一步增强(P<0.01)。结论痔疾洗液联合红光治疗对大鼠肛周创面模型术后有较好的治疗和改善作用,其作用机制可能与减轻机体炎症反应,促进皮肤愈合、再生有关。 Objective To observe the effect and mechanism of Hemorrhoid Lotion combined with red light therapy on postoperative recovery of perianal wound model in rats.Methods 60 Wistar rats were randomly divided into control group,model group,Hemorrhoid Lotion group,red light treatment group,Hemorrhoid Lotion combined with red light treatment group.After successful modeling each group was given corresponding treatment once a day for 14 consecutive days.The general condition of rats was observed,and the number of wound epithelialization and wound healing rate were measured.The expression and changes of interleukin-8(IL-8),tumor necrosis factor-α(TNF-α) transforming growth factor-β1(TGF-β1)and epidermal growth factor(EGF) in rat serum were detected by real-time fluorescence quantitative nucleic acid amplification(qPCR);the expression and changes of IL-8,TNF-α,TGF-β1 and EGF in rat skin were detected by enzyme linked immunosorbent assay(ELISA)in rat skin.Results Compared with model control group,the wound healing rates of Hemorrhoid Lotion group and red light treatment group were significantly increased(P<0.05),and the number of animals with wound epithelialization significantly enhanced(P<0.05),these effects were further enhanced in Hemorrhoid Lotion combined with red light treatment group(P<0.01);the expression of IL-8 and TNF-α was significantly up-regulated,while the expression of TGF-β1 and EGF was significantly down-regulated in model group(P<0.01);compared with model group,the expressions of IL-8 and TNFα in Hemorrhoid Lotion group and red light treatment group were significantly down-regulated(P<0.05),while the expression of TGF-β1 and EGF/p-EGFR was significantly up-regulated(P<0.05);the trend of IL-8,TNF-α,TGF-β1 and EGF/p-EGFR was further enhanced in Hemorrhoid Lotion combined with red light treatment group(P<0.01).Conclusion Hemorrhoid Lotion combined with red light therapy apparatus has a good therapeutic and improvement effect on perianal surgery in rats,and its mechanism may be related to reducing inflammatory reaction and promoting skin healing and regeneration.
