摘要
目的分析在人类精子冷冻过程中添加不同浓度的抗氧化冷冻保护剂MitoTEMPO(四甲基哌啶氮氧化物)对精子冷冻效果的影响。方法收集2019年7月—2019年11月体检来源精液206份,与患者沟通取得知情同意,检测精液的存活率、运动能力、DNA碎片等指标,随机分为不添加额外保护剂的自制精子冷冻保护剂空白对照组和添加高中低浓度自由基清除因子MitoTEMPO的实验组,冷冻后液氮中保存1周复苏,检测精液复苏后精子的各项指标,分析冷冻保护剂效果。将所有指标的测定结果,采用SPSS17.0数据分析系统,选择正确的统计学方法对数据资料进行整理、汇总、描述及分析冷冻保护剂效果。结果各组精子复苏后较冷冻前存活率和运动能力均降低(P<0.05),精子DNA碎片均升高(P<0.05),差异有统计学意义。三组添加MitoTEMPO组存活率、运动能力均比对照组高,其中高浓度组与对照组的差异有统计学意义(P<0.05)。三组添加MitoTEMPO组的DNA碎片均比对照组低,其中高浓度组和中浓度组与对照组的差异有统计学意义(P<0.05)。结论在精子冷冻中添加MitoTEMPO作为冷冻补充剂均具有提高精子冷冻效果的作用,可能与MitoTEMPO使低温处理中产生的氧化应激减低有关。
Objective To analyze the effect of adding different concentrations of anti-oxidant cryoprotectant MitoTEMPO(tetramethylpiperidine nitrogen oxide)during the freezing of human sperm on the freezing effect of sperm.Methods During July 2019 to November 2019,two hundred and six semen samples were included in this study communicate with patients to obtain informed consent,detect semen survival rate,motility,DNA fragmentation and other indicators,and randomly divide them into self-made sperm freezing without additional protective agent The protective agent blank control group and the experimental group added with high,medium and low concentrations of free radical scavenging factor MitoTEMPO were frozen and stored in liquid nitrogen for one week to thaw.The sperm indicators after thawing of the semen were detected and the effect of cryoprotectant was analyzed.The SPSS17.0 data analysis system was used to determine the results of all indicators,and the correct statistical methods were selected to sort,summarize,describe and analyze the effects of cryoprotectants.Results After thawing,the survival rate and motility of sperm in each group were lower than before freezing(P<0.05),and sperm DNA fragmentation was increased,the difference was statistically significant(P<0.05).The survival rate and exercise capacity of the three groups with MitoTEMPO added were higher than those of the control group,and the difference between the high concentration group and the control group was significant(P<0.05).The DNA fragments of the three groups with MitoTEMPO added were lower than those of the control group,and the difference between the high concentration group and the medium concentration group and the control group was significant(P<0.05).Conclusion The addition of MitoTEMPO as a freezing supplement in sperm freezing has the effect of improving the effect of sperm freezing,which may be related to the reduction of oxidative stress imposed by low temperature treatment by MitoTEMPO.
