阳和化岩汤干预乳腺癌癌前病变的效果及机制研究 被引量:6

摘要 目的探讨阳和化岩汤干预乳腺癌癌前病变的效果及机制。方法选取健康雌性未育SD大鼠110只,随机分为空白对照组、模型组、低剂量阳和化岩汤组、高剂量阳和化岩汤组和三苯氧胺组,每组22只。其中模型组、低剂量阳和化岩汤组、高剂量阳和化岩汤组和三苯氧胺组大鼠建立乳腺癌癌前病变模型,低剂量阳和化岩汤组、高剂量阳和化岩汤组和三苯氧胺组分别给予低剂量、高剂量阳和化岩汤和三苯氧胺,采用HE染色观察各组乳腺组织,免疫组化染色检测FGF-2和FIK-1蛋白表达,RT-PCR检测FGF-2和FIK-1 mRNA表达。结果模型组癌前病变、浸润性癌比例分别为66.67%(12/18)和27.78%(5/18),明显高于空白对照组(P<0.05);高剂量阳和化岩汤组癌前病变、浸润性癌比例分别为19.05%(4/21)和0.00%,较模型组降低(P<0.05);模型组FGF-2和FIK-1表达OD值、mRNA相对表达量分别为(0.892±0.121)和(0.913±0.112),(0.901±0.113)和(0.892±0.103),明显高于空白对照组(P<0.05);高剂量阳和化岩汤组FGF-2和FIK-1表达OD值、mRNA相对表达量分别为(0.640±0.103)和(0.722±0.100),(0.602±0.101)和(0.670±0.098),较模型组和剂量阳和化岩汤组降低(P<0.05),与三苯氧胺组比较差异无统计学意义(P>0.05)。结论阳和化岩汤抑制乳腺癌癌前病变,可能与调节FGF-2和FIK-1表达有关。 Objective To explore the effect and mechanism of Yanghe Huayan Decoction(阳和化岩汤)on breast precancerous lesions.Methods A total of 110 healthy female SD rats without pregnancy were randomly divided into blank control group,model group,Yanghe Huayan Decoction low and high dose groups and tamoxifen group,22 in each group.The model group,the Yanghe Huayan Decoction low and high dose groups and the tamoxifen group were used to establish the breast precancerous lesion model.The Yanghe Huayan Decoction low and high dose groups and tamoxifen group were given low and high dose of Yanghe Huayan Decoction and tamoxifen respectively.HE staining was used to observe the breast tissue of each group.The expressions of FGF-2 and FIk-1 were detected by immunohistochemistry and the expressions of FGF-2 and FIk-1 mRNA were detected by RT-PCR.Results The proportions of precancerous lesions and invasive carcinoma in the model group were 66.67%(12/18)and 27.78%(5/18),which were significantly higher than those in the blank control group(P<0.05).The proportions of precancerous lesions and invasive cancer in the Yanghe Huayan Decoction high dose group were 19.05%(4/21)and 0.00%,which were lower than those in the model group(P<0.05).In the model group,the OD value and relative mRNA expressions of FGF-2 and FIk-1 were 0.892±0.121,0.913±0.112,0.901±0.113 and 0.892±0.103,which were significantly higher than those in the control group(P<0.05).The OD value and mRNA expressions of FGF-2 and FIk-1 in the Yanghe Huayan Decoction high dose group were 0.640±0.103 and 0.722±0.100,0.602±0.101 and 0.670±0.098,which were lower than those in the model group and the Yanghe Huayan Decoction low dose group(P<0.05).There was no significant difference compared with those of the tamoxifen group(P>0.05).Conclusion Yanghe Huayan Decoction can inhibit precancerous lesions of breast cancer,which may be related to regulating the expressions of FGF-2 and FIk-1.
