健步汤对去势大鼠骨质疏松症模型骨密度及血清TNF-α、IL-6、IL-1表达的影响 被引量:8

摘要 目的:探讨健步汤对去势大鼠骨质疏松症模型骨密度及血清TNF-α、IL-6、IL-1的影响。方法:44只健康雌性SPF级SD大鼠,随机选取12只大鼠为假手术组,行剖腹手术但未摘除卵巢;其余32只大鼠全部行双侧卵巢切除术为手术组,喂养12周后,假手术组与手术组各随机处死2只行股骨HE染色镜下观察,并将其余40只大鼠行骨密度检测,检验造模全部成功后,将30只造模成功大鼠随机分为模型组、中药组和西药组,每组10只。西药组大鼠予戊酸雌二醇片和生理盐水配制成的混悬液灌胃,中药组大鼠予等效剂量健步汤灌胃,假手术组及模型组大鼠予等量生理盐水灌胃。连续给药12周后,检测大鼠腰椎股骨骨密度及血清TNF-α、IL-6及IL-1水平,光镜下观察股骨近端病理形态学改变。结果:模型组大鼠腰椎、股骨骨密度水平显著低于假手术组(P<0.05);中药组及西药组大鼠骨密度水平显著高于模型组(P<0.05),中药组与西药组大鼠腰椎、股骨骨密度比较,差异无统计学意义(P>0.05)。模型组大鼠血清TNF-α、IL-6和IL-1水平均明显高于假手术组,差异有统计学意义(P<0.05)。西药组与中药组大鼠血清TNF-α、IL-6和IL-1水平均低于模型组(P<0.05),中药组大鼠血清TNF-α、IL-6和IL-1水平与西药组比较,差异无统计学意义(P>0.05)。结论:健步汤可改善去势大鼠骨质疏松症模型的骨代谢程度,提高骨密度,其作用机制可能与降低血清中TNF-α、IL-6和IL-1的表达相关。 Objective: To investigate the effect of Jianbu Decoction on bone mineral density and serum TNF-α, IL-6 and IL-1 expression in ovariectomized rats with osteoporosis. Methods: A total of 44 healthy female SPF SD rats were randomly selected as sham operation group, and the remaining 32 rats were all treated with bilateral oophorectomy as operation group. After feeding for 12 weeks, 2 rats in sham operation group and operation group were randomly killed for observation under HE staining microscope, and the remaining 40 rats were tested for bone mineral density. After all the successful modeling, the remaining 30 rats were randomly divided into model group, Chinese medicine group and western medicine group. Western medicine group was given the suspension of estradiol valerate tablets and normal saline, Chinese medicine group was given the equivalent dose of Jianbu Decoction, and sham operation group and model group were given the same amount of normal saline. After 12 weeks of continuous administration, the bone mineral density of lumbar spine and femur,serum TNF-α, IL-6 and IL-1 expression in each group were observed and compared, and the pathological changes of proximal femur were observed under light microscope. Results: The bone mineral density of the lumbar spine and femur of the model group was significantly lower than that of the sham operation group(P<0.05). The bone mineral density of the rats in the Chinese medicine group and the western medicine group was significantly higher than that of the model group(P <0.05). There was no significant difference in bone mineral density of lumbar spine and femur between the Chinese medicine group and the western medicine group(P>0.05). The levels of serum TNF-α, IL-6 and IL-1 in the model group were significantly higher than those in the sham operation group, and the difference was statistically significant(P<0.05). The levels of serum TNF-α, IL-6 and IL-1 in the western medicine group and the Chinese medicine group were lower than those in the model group(P<0.05).There was no significant difference in serum TNF-α, IL-6 and IL-1 levels between the Chinese medicine group and the western medicine group(P >0.05). Conclusion: Jianbu Decoction can improve the degree of bone metabolism and increase bone density in ovariectomized rats with osteoporosis. Its mechanism of action may be related to reducing the expression of inflammatory factors such as TNF-α, IL-6 and IL-1 in serum.
