苗药血人参及其制剂对“气弱血弱体弱”冷病模型小鼠的作用研究 被引量:3

摘要 目的研究苗药血人参及其制剂对“气弱血弱体弱”冷病模型小鼠的作用。方法雄性KM小鼠,随机分为正常组、模型组、八珍颗粒阳性对照组、乙酸乙酯(YSYZ)部位高中低剂量组、芪胶升白胶囊高中低剂量组,每组10只。采用疲劳法+放血+泻下法复合造模方法。每天早上8点灌胃40%番泻叶水浸液,10点进行竭力游泳测试,下午2点给予相应药物灌胃,尾部放血0.2 g,隔2 d放一次,如此重复4周。每周观察小鼠体质量的改变;实验3周后,进行疲劳游泳时间的测定;第4周实验结束后眼球取血,ELISA法检测小鼠血浆ATP、cAMP、cGMP水平,并计算cAMP/cGMP比值。取脾、胸腺称重并计算脏器指数。结果与正常组比较,模型组小鼠皮毛稀疏且暗淡无光泽,饮食减少,鼠尾冰冷,体质量减轻,竭力游泳时间减小(P<0.01);与模型组比较,阳性药物组、YSYZ高剂量组、芪胶升白胶囊高剂量组小鼠身形匀称,活动自如,皮毛浓密、色乳白有光泽;各实验组脾指数明显高于正常组,除YSYZ部位中剂量组差异无统计学意义,其余各实验组差异有统计学意义(P<0.01或P<0.05);芪胶升白胶囊高剂量组脾指数高于模型组,其余各组脾指数均小于模型组,其中YSYZ部位中剂量组差异有统计学意义(P<0.01);与模型组比较,YSYZ高剂量组、芪胶升白胶囊高剂量组小鼠血清ATP、cAMP、cGMP含量降低(P<0.01或P<0.05)。结论苗药血人参及其制剂对“气弱血弱体弱”冷病模型小鼠具有一定的改善作用,其作用机制可能与调节小鼠ATP、cAMP、cGMP水平表达有关。 Objective To study the effect of Miao medicine blood ginseng and its preparations on mice with cold disease of weak qi,blood,and body.Methods Male KM mice were randomly divided into normal group,model group,Bazhen granule positive control group,high,medium and low-dose ethyl acetate group,high,medium and low-dose Qijiao Shengbai capsule group,with 10 mice in each group.The composite modeling method of fatigue+bleeding+purging was adopted.At 8:00 every morning,40%senna leaf water infusion was given to the stomach,and the swimming test was performed at 10:00.The corresponding drugs were given by gavage at 14:00,and 0.2 g of blood was bleeding from the tail,which was given once every 2 days,and the procedure was repeated for 4 weeks.The changes of body weight were observed every week.After 3 weeks of the experiment,the fatigue swimming time was measured.After the 4th week of the experiment,blood was taken from the eyeballs.The plasma ATP,cAMP and cGMP levels were detected by ELISA method,and the ratio of cAMP/cGMP was calculated.The spleen and thymus were weighed,and the organ index was calculated.Results Compared with the normal group,mice in the model group had sparse fur,reduced diet,cold tails,reduced body mass,and reduced exhaustive swimming time(P<0.01).Compared with the model group,the mice in the positive drug group,high-dose ethyl acetate group,and high-dose Qijiao Shengbai capsule group were well-proportioned and moved freely,with dense,creamy white and shiny fur.The spleen index of each experimental group was significantly higher than that of the normal group.The difference was not statistically significant in the middle-dose ethyl acetate group and the differences in the other experimental groups were statistically significant(P<0.01 or P<0.05).The spleen index of the high-dose Qijiaoshengbai capsule group was higher than that of the model group,and the spleen index of the other groups were lower than that of the model group.Among them,the difference in the middle-dose ethyl acetate group was statistically significant(P<0.01).Compared with the model group,the serum ATP,cAMP,and cGMP levels in the high-dose ethyl acetate group and the high-dose Qijiao Shengbai capsule group decreased,and the difference was statistically significant(P<0.01 or P<0.05).Conclusion Miao medicine blood ginseng and its preparations have a certain improvement effect on mice with cold disease,and its mechanism of action may be related to regulating the expression of ATP,cAMP,and cGMP.
