远志正丁醇提取物对SH-SY5Y细胞增殖及抗氧化损伤作用 被引量:1

Effects of n-butanol Extract from Polygala Tenuifolia on Proliferation and Antioxidant Damage of SH-SY5Y Cells

摘要 目的:研究远志正丁醇提取物对SH-SY5Y细胞的增殖作用及对H_(2)O_(2)诱导的SH-SY5Y细胞氧化损伤的保护作用。方法:增殖实验以0(对照组)、5、10、20、40、80和100 μg/mL的远志正丁醇提取物培养SH-SY5Y细胞48 h,观察细胞形态,用MTT法测算存活率。损伤实验重取细胞并随机分为空白组、对照组、实验组和损伤模型组。实验组以10、20和40 μg/mL的远志正丁醇提取物预培养实验组SH-SY5Y细胞24 h,观察细胞形态后,与模型组各加入H_(2)O_(2)至终浓度为150 μmol/L孵育,1 h后观察细胞的形态,测算存活率。结果:增殖实验中,100 μg/mL以下各组药物浓度均能促进正常SH-SY5Y细胞的增殖,5 μg/mL,10 μg/mL的药物浓度组与对照组相比,有显著性差异(P<0.05)。损伤实验中,与损伤模型组相比,各浓度组的细胞存活率有所上升,其中10 μg/mL浓度下的细胞存活率明显较高(P<0.05)。结论:远志正丁醇提取物在一定浓度内有促进SH-SY5Y细胞增殖及抗H_(2)O_(2)诱导的氧化损伤的功效。 Objective:To study the effect of n-butanol extract from Polygala tenuifolia on the proliferation of SH-SY5Y cells and its protective effect on oxidative damage induced by H_(2)O_(2).Methods:SH-SY5Y cells were cultured with 0(control group),5,10,20,40,80 and 100μg/mL n-butanol extract of Polygala tenuifolia for 48 h,cell morphology was observed and survival rate was measured by MTT method.In the injury experiment,the cells were randomly divided into blank group,control group,experimental group and injury model group.In the experimental group,SH-SY5Y cells were pre-cultured with 10,20 and 40μg/mL n-butanol extract from Polygala tenuifolia for 24 h.After observing the cell morphology,the SH-SY5Y cells in the model group were incubated with H_(2)O_(2)until the final concentration was 150μmol/L.Finally,the morphology of cells was observed and the survival rate was calculated.Results:In the proliferation experiment,the proliferation of normal SH-SY5Y cells was promoted by the concentration of drugs below 100μg/mL,there were significant differences between the drug concentration groups of 5μg/mL(P<0.05)and 10μg/mL(P<0.01).In the injury experiment,compared with the injury model group,the cell survival rate of each concentration group increased,and the cell survival rate at 10μg/mL(P<0.05)was significantly higher.Conlusion:The n-butanol extract from Polygala tenuifolia can promote the proliferation of SH-SY5Y cells and resist the oxidative damage induced by H_(2)O_(2).
