Prognostic Implication of Siglec-15 and CD8 Expressions in Endometrial Clear Cell Carcinoma

目的研究Siglec-15及CD8在子宫内膜透明细胞癌(ECCC)的表达,探讨2个标记物与患者总生存期(OS)及无进展生存期(PFS)的关系。方法回顾性分析27例ECCC术后患者的临床资料。免疫组织化学检测标本中Siglec-15及CD8的表达水平,以平均光密度表示,通过Pearson法分析2个标记物表达的相关性。根据平均光密度将27例患者分为高表达组和低表达组,通过Kaplan-Meier单因素分析法及Cox比例风险模型多因素分析法研究标本中Siglec-15及CD8的表达与患者OS及PFS的关系。结果Siglec-15及CD8在ECCC标本均有不同程度表达,Siglec-15表达水平与CD8表达水平呈负相关(P<0.005);Siglec-15高表达组病理级别更高(P<0.05),肌层浸润更深(P<0.005)。单因素分析显示,Siglec-15及CD8表达与患者的OS及PFS具有相关性;多因素分析显示,Siglec-15及CD8的表达并不是影响患者预后的独立因素。结论ECCC患者Siglec-15表达水平与CD8表达水平呈负相关;Siglec-15高表达的样本侵袭性更强;Siglec-15及CD8可能在影响患者预后方面共同起作用。

Objective To observe the expression of Siglec-15 and CD8 expressions in endometrial clear cell carcinoma and evaluate their prognostic value.Methods We conducted a retrospective study to quantify the IHC Siglec-15 and CD8 expressions in 27 primary endometrial clear cell carcinoma samples from a single cohort.The data were used to analyze the correlations of two markers in the specimens by Spearman method.In particular,the 27 patients were divided into high-expression group and low-expression group by median of average optical density,followed by their reciprocal combinations,including compartmental differences,and correlated them with overall survival(OS)and progression-free survival(PFS).Results The expressions of Siglec-15 and CD8 in endometrial clear cell carcinoma were detected.The expression of Siglec-15 was negatively correlated with CD8 in the specimens.The Siglec-15 high-expression cases tended to have deeper myometrial invasion and higher stage at presentation.The results of univariate analysis showed that both of Siglec-15 and CD8 expression were strongly relevant to the OS and PFS of endometrial clear cell carcinoma.In repeated Cox multivariable models adjusted by clinicopathologic factors,both of Siglec-15 and CD8 expression may not be independent adverse prognostic factor for the OS and PFS in all patients.Conclusion The expression of Siglec-15 was negatively correlated with the expression of CD8 in endometrial clear cell carcinoma.The samples with high Siglec-15 expression were more aggressive.The expression of Siglec-15 and CD8 may combine to affect the prognosis of patients Siglec-15 and CD8 may affect the prognosis of patients through interaction.
