决明子提取物对心肌梗死后心力衰竭大鼠心功能的影响及机制初探 被引量:3

摘要 目的研究决明子提取物对心肌梗死后心力衰竭大鼠心功能、炎症因子及心肌细胞凋亡和自噬水平的影响。方法通过结扎成年雄性SD大鼠前降支并饲养4周,成功制备心梗后心力衰竭模型42只,其中12只不做治疗处理,为对照组。另30只在给与脂多糖(lipopolysaccharide,LPS)刺激的基础上,即随机分为模型组+LPS组、决明子提取物高剂量+LPS组及低剂量+LPS组和假手术+LPS组,比较各组的心功能等指标。结果与其它各组相比,决明子高、低剂量组的心功能指标明显改善(P<0.05),炎症因子及N末端脑钠素原(NT-proBNP)、超敏肌钙蛋白(hs-TnT)水平显著降低(P<0.05),内质网应激相关蛋白(C/EBP-homologous protein,CHOP)、半胱胺酸蛋白酶蛋白12(Caspase-12)、半胱胺酸蛋白酶蛋白3(Caspase-3)表达水平降低,B淋巴细胞瘤-2(B-cell lymphoma-2,Bcl-2)、轻链3B蛋白(LC3B)表达水平升高(P<0.05)。病理HE染色结果显示,与其它各组相比,决明子提取物各给药组心肌梗死区域明显减少。结论决明子提取物可能通过抑制炎症反应,减轻心肌细胞凋亡,上调细胞自噬水平,在保护心肌细胞的基础上起到延缓心肌梗死后心力衰竭进展的作用。 Objective To study the effects of Semen Cassiae extracts on cardiac function,inflammatory factors,cardiomyocyte apoptosis and autophagy in rats with heart failure after myocardial infarction.Methods By ligating adult male SD rats and feeding them for 4 weeks,42 models of heart failure after myocardial infarction were successfully prepared,of which 12 were not treated as control group.The other 30 were randomly divided into model group+LPS group,cassia extract high-dose+LPS group,low-dose+LPS group and sham operation+LPS group on the basis of LPS stimulation,and the cardiac function and other indexes of each group were compared.Results Compared with other groups,the cardiac function indexes of cassia high and low dose groups were significantly improved(P<0.05),inflammatory factors and NT-proBNP,hs-TnT levels were significantly decreased(P<0.05),CHOP,Caspase-12,the expression level of Caspase-3 was decreased,and the expression levels of Bcl-2 and LC3 B were increased(P<0.05).The results of HE staining showed that the myocardial infarct area of each of the cassia seed extracts was significantly reduced compared with the other groups.Conclusion Cassia extract may inhibit the inflammatory response,reduce the apoptosis of cardiomyocytes,up-regulate the level of autophagy,and play a role in delaying the progression of heart failure after myocardial infarction.
