摘要
目的:研究光固化型纳米介孔二氧化硅负载甲硝唑复合水凝胶抗菌性及对牙髓干细胞(human dental pulp stem cells,hDPSCs)黏附性。方法:合成纳米介孔二氧化硅负载甲硝唑复合水凝胶(MSNs@gGel+MTR水凝胶),通过CCK-8和活/死细菌染色分析其对粪肠球菌(E.faecalis)和变形链球菌(S.mutans)的体外抗菌性。通过CCK-8分析该复合水凝胶对hDPSCs生物相容性,激光共聚焦显微镜和扫描电镜评估该复合水凝胶对hDPSCs黏附性能。结果:MSNs@gGel+MTR水凝胶对E.faecalis和S.mutans浮游细菌和细菌生物膜具有显著的抗菌性能。MSNs@gGel+MTR水凝胶对hDPSCs具有良好的生物相容性,可见hDPSCs黏附在MSNs@gGel+MTR水凝胶表面,且伸展良好;hDPSCs能迁移至MSNs@gGel+MTR组水凝胶内。结论:MSNs@gGel+MTR水凝胶具备抗菌和良好的hDPSCs黏附作用,可为牙髓再生的支架材料研究提供新思路。
Objective:The purpose of this study was to evaluate the antibacterial effect of metronidazole(MTR)loaded mesoporous silica nanoparticles(MSNs)-reinforced hydrogels scaffold(MSNs@gGel+MTR)in dental pulp regeneration.Methods:MSNs@gGel+MTR were synthesized by GelMA solution and photoinitiator,which containing MSNs loaded with metronidazole according to the previous study.The antimicrobial activity against E.faecalis and S.mutans was assessed by CCK-8 and live/dead-stained biofilms in vitro.Meanwhile,the biocompatibility and adhesion on human dental pulp stem cells(hDPSCs)was evaluated by CCK-8,confocal laser scanning microscopy(CLSM)and scaning electron microscopy(SEM)in vitro.Results:The CCK-8 and CLSM results showed the considerable antibacterial effect of MSNs@gGel+MTR on E.faecalis and S.mutans.Meanwhile,The CCK-8,CLSM and SEM results the good biocompatibility and cell adhesion of MSNs@gGel+MTR on hDPSCs.Conclusion:MSNs@gGel+MTR showed the good antibacterial effect and cell adhesion,which may be a promising approach in pulp regeneration in future.
