三子丸有效成分群对去卵巢骨质疏松大鼠骨代谢的影响 被引量:1

Effect of Integrated Effective Constituents from Sanziwan on Osteoporosis in Ovariectomized Rats

摘要 目的探讨三子丸有效成分群对去卵巢大鼠骨代谢的调控作用。方法4.5月龄的雌性SD大鼠去卵巢诱导骨质疏松症,灌胃给予阳性对照药尼尔雌醇(1 mg·kg^(-1),ig,每周1次)和三子丸有效成分群(15、30、60 mg·kg^(-1),ig,每天1次,每周6 d)12周。每周称量大鼠体质量;实验结束时分离大鼠子宫,称量质量;外周骨定量计算机断层扫描仪测量骨矿密度和骨矿含量;全自动生化分析仪测定血清磷、钙、肌酐、碱性磷酸酶(ALP)和抗酒石酸磷酸酶(TRAP);以Elisa试剂盒测定血清促肾上腺皮质激素(ACTH)、皮质酮、脱氧吡啶啉与肌酐比值(DPD/Cr)、骨保护素(OPG)的水平。结果与正常对照组比较,去卵巢模型组大鼠体质量显著增加,子宫质量显著降低,胫骨的骨密度和骨矿含量显著降低,血清钙、磷水平显著降低,血清DPD/Cr、TRAP和ALP水平升高,血清OPG水平降低,血清的ACTH和皮质酮显著增加。三子丸有效成分群对去卵巢大鼠的体质量没有显著的影响,可显著降低子宫质量,可增加去卵巢大鼠胫骨的骨密度和骨矿含量,提高血清磷、钙和OPG水平,降低血清DPD/Cr、TRAP、ACTH和皮质酮水平,对ALP活性没有显著影响。结论三子丸有效成分群通过抑制骨吸收减缓去卵巢大鼠的骨丢失。 OBJECTIVE To investigate the protective effects of integrated effective constituents of Sanziwan(SZW)on ovariectomy-induced bone loss.METHODS The 4.5-month-old female Sprague-Dawley rats were ovariectomized to develop osteoporosis,which was treated with vehicle,nylestriol(1 mg·kg^(-1),once a week)and SZW effective constituents(15,30,60 mg·kg^(-1),once a day,6 d per week)for 12 w,respectively.The body weights of rats were measured every week,while the uterus of rats were separated and weighted at the end of the experiment.Bone mineral density and bone mineral content were determined by peripheral quantitative computerized tomography densitometry.Serum phosphorus,calcium,adrenocorticotropic hormone(ACTH),corticosterone,deoxypyridinoline crosslinks to creatinine ratio(DPD/Cr),alkaline phosphate(ALP),tartrate-resistant acid phosphatase(TRAP),osteoprotegerin(OPG)levels were also measured.RESULTS Compared with the normal control group,the body weights of OVX rats increased significantly,the uterine weight,bone mineral density and bone mineral content of tibia were significantly decreased,the serum DPD/Cr,TRAP,ALP,ACTH and corticosterone levels were increased significantly,the serum phosphorus,calcium and OPG levels were significantly decreased.Treatment with SZW effective constituents significantly decreased the uterine weight,effectively increased the bone mineral density and bone mineral content of tibia in ovariectomized rats,without affecting the body weights,improved serum phosphorus,calcium,and OPG levels,lowered serum DPD/Cr,TRAP,ACTH,and corticosterone levels,but hardly changed ALP level in ovariectomized rats.CONCLUSION Through inhibiting bone resorption,SZW effective constituents has a definite protective effect on bone loss in ovariectomized rats.
