清热利湿方对特应性皮炎小鼠皮肤屏障蛋白表达的影响 被引量:2

摘要 目的探讨清热利湿方对特应性皮炎小鼠皮损及皮肤屏障蛋白表达的影响。方法将60只BALB/c雌性小鼠随机分为空白组、模型组、清热利湿方低剂量组、清热利湿方中剂量组、清热利湿方高剂量组、西替利嗪组,每组10只。除空白组外,其余组用卵清蛋白制备小鼠特应性皮炎模型。致敏结束后,清热利湿方低、中、高剂量组分别给予1 g/mL、2 g/mL、4 g/mL的清热利湿方灌胃,西替利嗪组给予25 mg/mL的西替利嗪灌胃,空白组、模型组灌胃等量的生理盐水,均1次/d,连续2周。灌胃结束后24 h内处死小鼠,取背部皮肤组织进行HE染色病理观察,RT-PCR检测小鼠皮肤组织中丝聚蛋白(FLG)、兜甲蛋白(LOR)、表皮蛋白(IVL)mRNA表达水平。结果临床表现及皮肤病理结果显示,各药物组小鼠皮损严重程度均轻于模型组,且清热利湿方低、中、高剂量组呈浓度梯度依赖性减轻。清热利湿方各组和西替利嗪组小鼠皮肤组织中FLG、LOR、IVL mRNA表达水平均明显高于模型组(P均<0.05),清热利湿方低、中、高剂量组各指标呈浓度梯度依赖性增高,清热利湿方高剂量组与西替利嗪组各指标比较差异均无统计学意义(P均>0.05)。结论清热利湿方可通过上调特应性皮炎小鼠皮肤屏障蛋白表达水平,减轻皮损。 Objective It is to observe the effect of Qingre Lishi Decoction on skin damage and skin barrier protein in atopic dermatitis mice.Methods Sixty BALB/c female mice were randomly divided into a blank group,a model group,a low-dose Qingre Lishi Decoction group,a medium-dose Qingre Lishi Decoction group,a high-dose Qingre Lishi Decoction group,and a cetirizine group,each group had 10 mice.Except for the blank group,the other groups used ovalbumin to prepare mouse atopic dermatitis models.After the sensitization,the low,medium and high dose groups of Qingre Lishi Decoction were respectively given 1 g/mL,2 g/mL,4 g/mL Qingre Lishi Decoction by gavage,and the cetirizine group was given 25 mg/mL cetirizine by gavage,the blank group and the model group were given the same amount of normal saline,all once a day for 2 consecutive weeks.The mice were sacrificed within 24 hours after the gavage,the back skin tissues were taken for HE staining pathological observation,and RT-PCR was used to detect the expression level of filaggrin(FLG),loricin(LOR)and involucrin(IVL)in the mouse skin tissues.Results Clinical manifestations and skin pathology results showed that the severity of skin lesions in the mice in each drug group was lighter than that in the model group,the lesions relieve in a concentration gradient-dependent manner in the low,medium and high dose groups of Qingre Lishi Decoction.The expression levels of FLG,LOR,and IVL mRNA in the skin tissues of mice in the Qingre Lishi Decoction groups and the cetirizine group were significantly higher than those in the model group(all P<0.05),the indexes increased in a concentration gradient-dependent manner in the low,medium and high dose groups of Qingre Lishi Decoction,there was no statistically significant difference in the indexes between the high-dose Qingre Lishi Decoction group and the cetirizine group(all P>0.05).Conclusion Qingre Lishi Decoction can up-regulate the expression level of skin barrier protein in mice with atopic dermatitis and reduce skin damage.
