摘要
为了构建绵羊附睾上皮细胞氧化应激模型,试验使用H_(2)O_(2)刺激绵羊附睾上皮细胞,并检测细胞凋亡率,测定细胞内活性氧(ROS)、丙二醛(MDA)含量以及细胞内还原型谷胱甘肽/氧化型谷胱甘肽(GSH/GSSG)比值,同时测定培养液内乳酸脱氢酶(LDH)含量。结果表明:当培养液内添加终浓度为400μmol/L的H_(2)O_(2)时,与不添加H_(2)O_(2)的对照组相比,细胞凋亡率升高,细胞内ROS、MDA含量上升,细胞内GSH/GSSG比值降低,培养液内LDH含量升高。说明本研究成功构建了绵羊附睾上皮细胞氧化应激模型。
In order to establish the oxidative stress model of sheep epididymal epithelial cells,H_(2)O_(2)was used to stimulate sheep epididymal epithelial cells,and the apoptosis rate,the content of reactive oxygen species(ROS),malondialdehyde(MDA),the ratio of reduced glutathione/oxidized glutathione(GSH/GSSG)and the content of lactate dehydrogenase(LDH)in the culture medium were detected.The results showed that when the final concentration of H_(2)O_(2)was 400μmol/L,compared with the control group without H_(2)O_(2),the apoptosis rate increased,the content of ROS and MDA increased,the ratio of GSH/GSSG decreased,and the content of LDH increased.The results showed that the oxidative stress model of sheep epididymal epithelial cells was successfully constructed.
