摘要
目的研究生长抑制因子蛋白家族成员4 (inhibitor of growth family memember 4,ING4)、p53和p21在子宫内膜样癌中的表达及意义。方法采用组织芯片技术和免疫组织化学方法检测正常子宫内膜组(正常子宫内膜组织20例)、不典型增生子宫内膜组(不典型增生子宫内膜组织30例)和子宫内膜样癌组(子宫内膜样癌组织60例)中ING4、p53和p21的蛋白表达情况及其与临床病理特征之间的关系;同时采用Western blot法比较20例子宫内膜样癌组织及对应癌旁正常子宫内膜组织中ING4、p53和p21蛋白表达水平。结果子宫内膜样癌组中ING4和p21的阳性表达率低于不典型增生子宫内膜组和正常子宫内膜组,差异均具有统计学意义(均P<0.05)。子宫内膜样癌组中p53的阳性表达率高于不典型增生子宫内膜组和正常子宫内膜组(均P<0.05)。在不同病理分期、分化程度和肌层浸润的子宫内膜样癌患者组织中ING4、p53和p21蛋白的表达水平比较,差异均具有统计学意义(均P<0.05)。Western blot法结果显示,癌旁正常子宫内膜组织中ING4和p21蛋白表达水平均高于子宫内膜样癌组织(均P<0.05),p53在子宫内膜样癌组织中的表达水平高于癌旁正常子宫内膜组织(P<0.05)。结论 ING4及p21的表达在子宫内膜样癌中降低,p53表达则升高。ING4、p53及p21的表达与子宫内膜样癌病理分期、分化程度和肌层浸润有关。ING4-p53-p21信号通路在子宫内膜样癌中发挥重要作用。
Objective To study the expression and significance of inhibitor of growth family member 4(ING4),p53 and p21 in endometrioid carcinoma.Methods This study enrolled 20 samples of normal endometrial tissues(normal endometrial group),30 samples of dysplasia endometrial tissues(dysplasia endometrial group),and 60 samples of endometrioid carcinoma tissues(endometrioid carcinoma group).The tissue chip technique and immunohistochemical method were used to detect the protein expressions of ING4,p53 and p21 in the three groups,and their relationship with clinicopathological characteristics were analyzed.Western blot was used to compare the expression levels of ING4,p53 and p21 in 20 samples of endometrioid carcinoma tissues and the corresponding adjacent normal endometrial tissues.Results The positive expression rates of ING4 and p21 in the endometrioid carcinoma group were significantly lower than those in the normal endometrial and dysplasia endometrial groups,and the differences were statistically significant(all P<0.05).The positive expression rate of p53 in the endometrioid carcinoma group was significantly higher than those of the normal endometrial and dysplasia endometrial groups(both P<0.05).There were statistically significant differences in the expression levels of ING4,p53 and p21 in tissues of endometrioid carcinoma patients with different pathological stages,degree of differentiation,and depth of myometrial invasion(all P<0.05).Western blot results showed that the levels of ING4 and p21 in the normal endometrial tissues adjacent to cancer were significantly higher than those in the endometrioid carcinoma tissues(both P<0.05),and the level of p53 in endometrioid carcinoma tissues was higher than that in the normal endometrial tissues adjacent to the cancer(P<0.05).Conclusions The levels of ING4 and p21 are significantly reduced in endometrioid carcinoma, while the expression of p53 was increased. The expressions of ING4, p53 and p21 are negatively correlated withthe degree of differentiation, pathological stage and depth of myometrial invasion. ING4-p53-p21 signaling pathway plays an important rolein endometrioid carcinoma.
