缺氧在小鼠肺间质肌成纤维细胞化过程中的作用及黄芩苷的保护机制 被引量:1

摘要 目的研究缺氧在小鼠肺间质肌成纤维细胞化进程中的作用及可能机制,同时评价黄芩苷的保护作用。方法选择6周龄清洁级别小鼠共30只,随机分成三组,分别为常氧组、低氧组、低氧+黄芩苷组。低氧组和低氧+黄芩苷组每天均需进入11%氧浓度低氧氧舱8 h,每周入舱6 d。4周后取小鼠肺组织,通过免疫组化技术分析三组小鼠肺间质胶原蛋白Ⅰ、胶原蛋白Ⅲ及α平滑肌肌动蛋白(α-SMA)的表达的差异,通过Western blot技术检测肺组织中α-SMA表达差异。结果三组间小鼠肺间质胶原蛋白Ⅰ、胶原蛋白Ⅲ、α-SMA以及肺组织α-SMA的表达量比较,差异均有统计学意义(F分别=75.04、15.10、29.71、264.48,P均<0.05)。低氧组小鼠肺间质胶原蛋白Ⅰ、胶原蛋白Ⅲ、α-SMA以及肺组织α-SMA表达量均明显高于常氧组(t分别=11.74、4.69、7.67、22.04,P均<0.05),低氧+黄芩苷组干预后小鼠肺间质胶原蛋白Ⅰ、胶原蛋白Ⅲ、α-SMA以及肺组织α-SMA的表达量均明显低于低氧组,差异均有统计学意义(t分别=5.12、3.02、4.51、13.40,P均<0.05)。结论缺氧导致肺间质肌成纤维细胞化进程相关的胶原蛋白Ⅰ、胶原蛋白Ⅲ、α-SMA指标明显上调,提示缺氧参与这一进程。黄芩苷干预后这些指标明显下调,提示具有保护性作用。 Objective To study the role and possible mechanism of hypoxia in the process of myofibroblast transdifferentiation of lung interstitium in mice and evaluate the protective effect of baicalin.Methods A total of 30 clean grade mice with 6-week-old were randomly divided into three groups,namely the normoxia group,the hypoxia group,and the hypoxia+baicalin group.The hypoxia group and the hypoxia+baicalin group received 11% oxygen concentration hypoxic chamber for 8 hours every day,6 days a week.Four weeks later,immunohistochemistry was used to detect the expressions of collagen Ⅰ,collagen Ⅲ and α-SMA in lung interstitium of mice among three group,and western blot was used to detect the difference inα-SMA expression in lung tissues.Results The expression levels of collagen Ⅰ,collagen Ⅲ,α-SMA in lung interstitium of mice and α-SMA in lung tissue were significantly different among the three groups(F=75.04,15.10,29.71,264.48,P<0.05).The expression levels of collagen Ⅰ,collagenⅢ,α-SMA in lung interstitium of mice andα-SMA in lung in hypoxic group were significantly higher than those in normoxic group(t=11.74,4.69,7.67,22.04,P<0.05).The expression levels of collagen Ⅰ,collagen Ⅲ,α-SMA in lung interstitium of mice andα-SMA in lung after baicalin intervention in the hypoxia+baicalin group were significantly lower than those in hypoxia group(t=5.12,3.02,4.51,13.40,P<0.05).Conclusion Hypoxia significantly up-regulates the levels of collagen Ⅰ,collagen Ⅲ,α-SMA related to the process of myofibroblasts in the lung interstitium.But these indexes significantly downregulate after baicalin intervention,suggesting a protective effect.
