摘要
为了探究chi-miR-128-3p和chi-miR-218对钟基因Nr1d2的靶向调控关系,以psiCHECK TM-2为载体,构建了野生型和突变型载体,转染293A细胞系,并通过双荧光素酶系统检测,chi-miR-218 mimics能使野生型荧光素酶活性极显著下降,而对突变型荧光素酶活性无影响,说明chi-miR-218靶向Nr1d2基因。该结果对研究受环境影响的生物节律如毛囊发育等现象的机制具有重要意义。
To investigate the regulation of biorhythmic genes Nr1d2 targeted by chi-miR-128-3p and chi-miR-218,wild-type and mutant vectors were constructed successfully with psiCHECKTM-2 and transfected into 293A cell line.The target relationships between chi-miR-128-3p or chi-miR-218 and the Nr1d2 gene were determined by a dual luciferase assays system.The chi-miR-218 mimics could significantly reduce wild-type luciferase activity,but had no effect on mutant luciferase activity.It indicated that chi-miR-218 targeted to the Nr1d2 gene.This result is of significant meaning to the further study of the mechanism of environmental rhythm phenomena such as hair follicle development.
