肾透明细胞癌低氧相关基因预后模型的建立与应用

Development and application of a hypoxia-related gene prognosis model for clear cell renal cell carcinoma

摘要 [目的]低氧是肾透明细胞癌内肿瘤微环境的重要组成部分,在肿瘤的发生与发展中起着至关重要的作用。利用癌症基因组图谱(TCGA)数据库建立低氧相关基因模型。[方法 ]提取低氧相关基因的表达进行单因素和多因素Cox回归分析建立模型。分析模型与临床分期以及免疫细胞浸润的相关性。[结果]构建5个低氧相关基因模型(ISG20、PLIN2、TMEM45A、PCK1和ENO2)。KM曲线显示高风险组的患者预后更差(P <0.001)。ROC曲线显示1a、3a和5a的曲线下面积(AUC)分别为0.703、0.690和0.693。[结论]该研究为肾透明细胞癌患者建立了一个稳健的低氧相关基因模型,其可作为肾透明细胞癌的潜在预后生物标记物。 [Objective]Hypoxia is an essential part of the tumor microenvironment in clear cell renal cell carcinoma(ccRCC),which plays a vital role in tumor development and progression. The hypoxia-related gene model was established by using The Cancer Genome Atlas(TCGA) database. [Method]The expression of hypoxia-related genes were extracted and analyzed by Cox regression analysis. The correlation of the model with clinical stage and immune cell infiltration was analyzed.[Result]Five hypoxia-related gene models(ISG20,PLIN2,TMEM45 A,PCK1 and ENO2) were constructed. The KM curve showed that the prognosis of the high-risk group was worse(P < 0. 001). The area under the curve(AUC) of 1 year,three years,and five years was 0. 703,0. 690,and 0. 693.[Conclusion]The study established a robust hypoxia-related gene model for patients with renal clear cell carcinoma,which can be used as a potential prognostic biomarker for renal clear cell carcinoma.
