天麻素对SNL模型大鼠脊髓背角内HDAC1-5、pERK、p-p38及pJNK的影响

Effects of gastrodin on HDAC1-5,pERK,p-p38 and pJNK in the spinal dorsal horn of SNL model rats

摘要 [目的]观察天麻素对SNL模型大鼠脊髓背角内HDAC1-5、pERK、p-p38及pJNK的影响。[方法]选取健康雄性SD大鼠48只,随机数字表法分成正常组、假手术组、SNL组及天麻素组,每组12只。Von-Frey细丝测量大鼠足底痛行为,免疫荧光染色观察大鼠脊髓背角内HDAC1-5的表达位置,Western Blot法定量分析脊髓背角内HDAC1-5、pERK、pJNK及p-p38 MAPK的表达水平。[结果]假手术-生理盐水组大鼠术侧后足痛阈与正常组相比无显著差异(P>0.05),SNL-生理盐水组大鼠术侧后足痛阈显著降低(P <0.05);给予天麻素腹腔注射治疗后大鼠术侧后足痛阈明显增加(P <0.05)。与假手术-生理盐水组相比,SNL-生理盐水组HDAC1-5、pERK、p-p38及pJNK的表达明显增加,差异具有统计学意义(P <0.05);腹腔注射给予天麻素可观察到HDAC1-5、pERK、p-p38及pJNK的表达得到显著降低,具有统计学差异(P <0.05)。[结论]天麻素抑制脊髓背角内组蛋白去乙酰化酶1-5的表达及pERK、p-p38及pJNK信号通路的激活,增加了大鼠术侧后足痛阈,且效果维持到停药后2 w左右,显著缓解SNL诱导的慢性神经病理性痛。 [Objective]To observe the effects of gastrodin on HDAC1-5,pERK,p-p38 and pJNK in the spinal dorsal horn of SNL model rats.[Method]Forty-eight healthy male SD rats were selected and divided into normal group,sham operation group,SNL group and gastrodin group by random number table,with 12 rats in each group. Von-Frey filaments measured the behavior of plantar pain in rats,immunofluorescence staining was used to observe the expression position of HDAC1-5 in the rat spinal dorsal horn,and Western Blot was used to quantitatively analyze the expression level of HDAC1-5,pERK,pJNK and p-p38 in the spinal dorsal horn.[Result]Compared with the normal group,the pain threshold of the sham-operated-saline group was not significantly different from that of the normal group(P > 0. 05),and the pain threshold of the foot of the SNL-saline group was significantly lower(P < 0. 05);Given after intraperitoneal injection of gastrodin,the posterior foot pain threshold of rats was significantly increased(P < 0. 05). Compared with the sham operation-saline group,the expressions of HDAC1-5,pERK,p-p38 and pJNK in the SNL-saline group were significantly increased,with statistical differences(P <0. 05);intraperitoneal injection of gastrodin can be observed. The expressions of HDAC1-5,pERK,p-p38 and pJNK were significantly reduced,with statistical differences(P < 0. 05). [Conclusion]Gastrodin can inhibit the expression of histone deacetylase 1-5 and the activation of pERK,p-p38 and p INK signaling pathways in the spinal dorsal horn,increase the pain threshold of rats’ postoperative foot,and the effect can be maintained until about 2 weeks after drug withdrawal,which can significantly alleviate the chronic neuropathic pain induced by SNL.
