鱼皮低聚肽对小鼠急性酒精性肝损伤的保护作用 被引量:5

摘要 目的:探究鱼皮低聚肽(FOPs)对酒精损伤小鼠肝功能及糖脂代谢的影响。方法:将60只KM小鼠作为研究对象分为空白组(蒸馏水)、模型组(蒸馏水)、阳性组(水飞蓟素,200 mg/kg BW)、FOPs组(250、500、1000 mg/kg BW)。实验第33天灌胃制ALD模型。对小鼠各相关联指标(肝功能、血糖、血脂和氧化)的含量和活性进行检测,并量化分析肝组织的脂滴情况进行。结果:FOPs氨基酸的成分组成甘氨酸(Gly)含量最高,其中90.61%均为低于1000 U的小肽;FOPs各剂量组小鼠血清的肝功能指标AST、ALT活力和血脂TG、TC含量显著降低(P<0.05),250、500 mg/kg BW FOPs的TC含量略低于阳性组;FOPs各剂量组与模型组比较,肝脏合成蛋白指标TP、ALB含量随浓度的增加而升高,FOPs(250、1000 mg/kg BW)组血糖GLU含量则明显增高(P<0.05);250、1000 mg/kg BW剂量组肝组织过氧化指标MDA含量显著降低(P<0.05),FOPs各剂量组肝组织抗氧化指标GSH活性明显增高,甚至优于空白组,与模型组相比差异显著(P<0.05)。小鼠肝组织油红O染色表明,FOPs缓解小鼠急性酒精肝细胞损伤的脂肪病变程度显著(P<0.05)。结论:FOPs在急性乙醇小鼠的肝功能损伤和糖脂代谢方面方面具有良好的保护功能。 Objective To investigate the effects of Fish skin oligopeptides(FOPs)on liver function and glucose and lipid metabolism in mice with alcohol injury.Method Totally 60 KM mice were divided into blank(distilled water),model(distilled water),positive group(silymarin,200 mg/kg BW)and FOPs groups(250,500,1000 mg/kg BW).On the 33rd day,the ALD model was made by gavage.The content and activity of the relevant indicators(liver function,blood sugar,blood lipid and oxidation)of the mice were detected,and the lipid droplets in the liver tissue were quantitatively analyzed.Result The contents of glycine(Gly)in FOPS amino acids were the highest,90.61%of which were small peptides less than 1000 U.The activities of AST and ALT and the contents of TG and TC in serum of mice in FOPs were significantly decreased(P<0.05),The content of TC in 250 and 500 mg/kg BW FOPs was slightly lower than that of positive group.Compared with model group,the contents of TP and Alb in FOPs increased with the increase of concentration,while the content of GLU in 250 and 1000 mg/kg BW groups was significantly increased(P<0.05).The content of MDA in liver tissue in 250 and 1000 mg/kg BW groups was significantly decreased(P<0.05),while the activity of GSH in FOPS groups was significantly higher,even better than that of blank group,and the difference was significant compared with model group(P<0.05).Oil red O staining of mice liver tissue showed that FOPs significantly alleviated the degree of fatty lesions in mice with acute alcoholic liver cell injury(P<0.05).Conclusion FOPS has a good protective effect on liver function injury and glycolipid metabolism in acute ethanol mice.
