人参茎叶皂苷和硒溶液(GSe)增强猪伪狂犬病疫苗免疫反应 被引量:1

Immunopotentiating effect of ginseng stem-leaf saponins and selenium(GSe)on pseudorabies virus vaccine

摘要 为观察含人参茎叶皂苷(GSLS)和硒(Se)的溶液(GSe)对猪伪狂犬病(PR)疫苗免疫的影响,本研究给小鼠免疫PR疫苗前注射含GSLS和Se的灭菌溶液,测定免疫后小鼠血清中特异性gB抗体及其亚类水平,淋巴细胞增殖能力,淋巴细胞分泌细胞因子(IFN-γ、IL-2、IL-4、IL-6和IL-10)水平以及猪伪狂犬病病毒(PRV)攻毒后小鼠的存活率,并观察GSe对巨噬细胞RAW 264.7的影响。结果表明,与对照组比较,在PR疫苗免疫前注射GSe可显著提高gB抗体及其亚类IgG1和IgG2a水平、淋巴细胞增殖水平、细胞因子(IFN-γ、IL-2、IL-4、IL-6和IL-10)和小鼠对PRV攻毒的抵抗力。同时,GSe提高了巨噬细胞分泌IL-6、IL-10和TNF-α水平以及细胞表面共刺激分子CD40、CD80和CD86的表达水平。因此,GSe可通过增强动物的固有免疫提高对PR疫苗的免疫应答。 To determine the immunoenhancement effect of GSe solution containing ginseng stemleaf saponins(GSLS)and selenium(Se)on the pseudorabies(PR)vaccine,mice were injected with sterile GSe solution containing GSLS and Se at pre-immunization of PR vaccine(GSe/pre-PRV).Results showed GSe/pre-PRV induced significantly higher serum gB-specific antibody responses(IgG,IgG1and IgG2a),increased lymphocyte proliferation,induced higher production of cytokines(IFN-γ,IL-2,IL-4,IL-6and IL-10)by splenocytes,improved the survival rate of immunized mice challenged with PRV.Additionally,GSe could improve the IL-6,IL-10and TNF-αproductions as well as the costimulatory molecules CD40,CD80and CD86expression of macrophages.In conclusion,GSe can enhance the immune responses of mice to the PRV vaccine by improving the innate immune responses.
