摘要
为探究围产期酮病奶牛极低密度脂蛋白(VLDL)的分泌特征,本试验利用ELISA试剂盒检测酮病奶牛血液中VLDL-ApoB的含量,超速离心分离VLDL颗粒,透射电镜观察颗粒大小,同时利用ELISA试剂盒以及LC-MS组学技术对奶牛血液VLDL颗粒进行脂质组学检测。结果显示,临床型酮病奶牛血液中VLDL-ApoB的含量显著低于健康对照奶牛,且透射电镜结果显示,临床型酮病奶牛血液VLDL颗粒较健康对照奶牛出现异常变大情况。不同脂质含量分析结果显示,奶牛酮病发生过程中磷脂(PL)出现显著或极显著的降低。结果表明,能量负平衡导致奶牛酮病发生过程中ApoB以及PL含量均受到抑制且该因素进一步促进疾病进程。
In order to investigate the characteristics of VLDL secretion in perinatal ketotic dairy cows,the content of VLDL-ApoB in blood of ketotic dairy cows was detected by ELISA kit,the particle size of VLDL particles was observed by transmission electron microscope after ultracentrifugation,and the lipid histology of VLDL particles in blood of dairy cows was detected by ELISA kit and LC-MS histology technology.The results showed that the content of VLDL-ApoB in the blood of clinical ketotic cows was significantly lower than that of the healthy control cows,and the results of transmission electron microscope showed that the blood VLDL particles of clinical ketotic cows were abnormally larger than those of the healthy control cows.The results of different lipid contents showed that phospholipids decreased significantly during the occurrence of ketosis.The results showed that ApoB and phospholipids contents were inhibited due to negative energy balance during the occurrence of ketosis,which promoted the disease process.
