摘要
通过构建国家果树种质沈阳山楂圃内保存的48份资源的分子身份证,以探索建立山楂资源分类鉴定的分子技术体系。利用14对SSR引物对山楂资源进行扩增,每对引物检测出2~4个等位基因,平均为2.14个。各引物的Shannon’s多样性指数I变幅为0.283~0.686,均值为0.582;多态信息含量(PIC)变幅为0.139~0.372,均值为0.311。遗传分化系数Fst变幅为0.057~0.742,均值为0.447,反映出群体间存在较大的遗传分化。山楂资源间的相似系数为0.290~0.968,其中羽裂山楂与伏山楂亲缘关系较近;羽裂山楂大果变种与湖北山楂的亲缘关系较近;毛山楂与辽宁山楂的亲缘关系最近,与其他种山楂亲缘关系较远。将14对SSR引物获得的相应等位基因赋值编码,串联构建山楂资源的分子身份证。结果表明SSR标记技术可作为山楂资源鉴定和分子身份证构建的有效技术手段,从而提高山楂资源鉴定、评价和利用效率。
To explore a molecular technology system for the classification and identification of Crataegus germplasm resources,the molecular ID of 48 Crataegus resources preserved in the National Repository of Hawthorn Germplasm Resources in Shenyang were constructed.Forteen pairs of SSR markers were used to amplify the Crataegus resources.The number of alleles per pair of primers ranged from 2 to 4,averagely 2.14 alleles per locus.Shannon’s information index ranged from 0.283 to 0.686 with an average of 0.582;Polymorphic information content(PIC)ranged from 0.139 to 0.372 with an average of 0.311 per locus.Fst ranged from 0.057 to 0.742 with an average of 0.447,indicating a large genetic differentiation among populations.Similarity coefficient among Crataegus resources ranged 0.290 to 0.968.Genetic relationship between C.pinnatifida and C.brettschneideri was closer,C.pinnatifida.var and C.hupehesis had similar situations.Compared with these Crataegus species,C.maximowiczii and C.sanguinea had closer relationship.The corresponding alleles achieved from 14 pairs of SSR markers were assigned and coded,then the character strings were applied to construct molecular ID of Crataegus resources.The results indicated that SSR marker can be used as an effective technique for the variety identification and molecular ID construction of Crataegus resources.Molecular ID could improve the identification,evaluation and utilization efficiency of Crataegus germplasm resources.
