补中益气汤对实验性结肠炎Treg细胞的影响

Effect of Buzhong Yiqi decoction on Treg cells of experimental colitis

目的:观察补中益气汤对实验性结肠炎Treg细胞的影响。方法:将SPF级雄性SD大鼠48只随机分为空白对照组、模型组、补中益气汤低、中、高剂量组和美沙拉嗪组,每组各8只。采用5%葡聚糖硫酸钠溶液制备UC大鼠模型。造模结束后,补中益气汤组分别予低、中、高剂量补中益气汤灌胃,空白对照组和模型组给予生理盐水灌胃,美沙拉嗪组给予美沙拉嗪溶液灌胃,连续7天。观察大鼠结肠组织形态学改变,检测大鼠外周血清中的IL-10的含量、结肠组织中Foxp3基因表达水平及Foxp3蛋白的含量,并检测大鼠外周血、肠系膜淋巴结、结肠固有层和脾脏中Treg细胞的数量。结果:①与模型组比较,补中益气汤中、高剂量组和美沙拉嗪组均可上调大鼠血清中IL-10含量(P<0.01);②与模型组比较,补中益气汤低、高剂量组和美沙拉嗪组结肠组织中Foxp3的基因表达均有所升高(P<0.05),各给药组大鼠结肠中的Foxp3蛋白表达均明显增加(P<0.01);③模型组大鼠结肠固有层中的Treg细胞数量低于各组,且与补中益气汤中剂量组和美沙拉嗪组比较,差异有统计学意义(P<0.01)。结论:补中益气汤可以改善DSS诱导的大鼠结肠黏膜损伤,其作用机制可能与上调损伤部位Treg细胞数量,促使Treg分泌IL-10有关。

Objective:To observe the effect of Buzhong Yiqi decoction on Treg cells of experimental colitis.Methods:48 male SD rats of SPF grade were randomly divided into blank control group,model group,low-,mediumand high-dose Buzhong Yiqi decoction groups and maisalazine group,8 rats in each group.using 5%DSS solution to make UC rat model.After the molding was finished,Buzhong Yiqi decoction groups were respectively given low-,medium-and high-dose Buzhong Yiqi decoction by gavage,blank control group and model group were given normal saline by gavage,mesalazine group was given mesalazine solution by gavage,continuously for 7 days.The histomorphological change of colon in rats were observed.The indexes in rats were detected such as the content of IL-10 in peripheral serum of rats,the level of gene expression of Foxp3 and the protein content of Foxp3 in colonic tissue of rats as well as the number of Treg cells in peripheral blood,mesenteric lymph node,colonic lamina propria and spleen of rats.Results:1.Compared with the model group,the levels of IL-10 in serum of rats in medium-and high-dose Buzhong Yiqi decoction groups and mesalazine group were significantly increased(P﹤0.01);2.Compared with model group,in low-and high-dose Buzhong Yiqi decoction groups and mesalazine group,the gene expression of Foxp3 in colonic tissue of rats was somewhat increased(P﹤0.05);while in each administration group,the protein expression of Foxp3 in colonic tissue of rats was significantly increased(P﹤0.01).3.In the model group,the number of Treg cells in colonic lamina propria of rats was lower than that in other each group,and the difference was statistically significant compared with medium-dose Buzhong Yiqi decoction group and mesalazine group(P﹤0.01).Conclusion:Buzhong Yiqi decoction can improve DSS-induced colonic mucosal injury of rats.Its mechanism may be related to upregulating the number of Treg cells of injured part and promoting Treg to secrete IL-10.
