摘要 目的研究CD248的全人源单克隆抗体IgG78对HFL-1人胚肺成纤维细胞生物学功能的影响。方法表达和纯化CD248人源性抗体IgG78,用IgG78处理HFL-1细胞后,利用CCK-8实验检测细胞增殖,Transwell^(TM)实验检测细胞迁移,流式细胞术检测细胞凋亡和细胞周期,Western blot法检测CD248及Ⅰ型胶原蛋白α1(COL1A1)和α平滑肌肌动蛋白(α-SMA)的表达变化。结果IgG78可有效抑制HFL-1细胞的增殖和迁移,诱导细胞凋亡,并引起细胞周期的G1期阻滞。IgG78还可抑制HFL-1细胞中CD248及纤维化相关蛋白COL1A1和α-SMA的表达。结论CD248抗体IgG78可抑制成纤维细胞的生物学活性及纤维化相关蛋白的表达,该抗体对CD248特异性增高的肿瘤或纤维化疾病的治疗有潜在应用价值。 Objective To investigate the effect of CD248 specific antibody IgG78 on the biological function of HFL-1 human fetal lung fibroblasts.Methods HFL-1 cells were treated with human antibody IgG78 expressed and purified.Cell proliferation was detected by CCK-8 assay,cell migration was detected by Transwell^(TM) assay,cell apoptosis and cell cycle were detected by flow cytometry,and the expressions of CD248,collagen typeⅠalpha 1(COL1A1)and α-smooth muscle actin(α-SMA)were detected by Western blotting.Results IgG78 effectively inhibited the proliferation and migration of HFL-1 cells,induced apoptosis,and arrested cell cycle at G1 phase.IgG78 also inhibited the expression of CD248 and fibrosis-related proteins COL1A1 and α-SMA in HFL-1 cells.Conclusion CD248 specific antibody IgG78 can inhibit the biological activity of fibroblasts and the expression of fibrosis-related proteins,indicating that it can be used in the treatment of tumors and fibrotic diseases with specific increase in CD248 expression.
