摘要
目的:观察直肠癌组织中miR-93表达水平与直肠癌组织血管生成参数及动态增强磁共振血流灌注(DCE-MR)参数的相关性,为miR-93在直肠癌血管生成中的评价意义及价值提供研究基础。方法:随机选取2018年04月至2019年04月经术后病理确诊为直肠癌的患者32例,所有患者术前均行直肠DCE-MR检测,对肿瘤横断面兴趣区进行K^(trans)、K^(ep)、V_(e)和iAUC的定量测量,术后病理以Western blot检测血管内皮细胞生长因子(vasular endothelial growth factor,VEGF)表达,以免疫组化法检测微血管密度(microvessel density,MVD),以qRT-PCR检测miR-93在直肠癌组织及配对癌旁直肠组织中的表达,以Pearson rank相关性检验观察miR-93表达水平与直肠癌直径、直肠癌组织血管生成参数(VEGF及MVD)、磁共振成像非形态学表征(ADC及TIC分型)及直肠MRI血流灌注参数(K^(trans)、K^(ep)、V_(e)及iAUC)的相关性。结果:miR-93在直肠结论:miR-93表达水平与直肠肿瘤组织中血管生成参数及磁共振血流灌注参数具有相关性,可能用来对直肠癌血管生成进行评估,并作为直肠癌生物学行为的预测参数。
Objective:To observe miR-93 expression in rectal cancer tissues and the correlation between miR-93 and tumor angiogenesis dates or DCE-MR blood perfusion dates,to provide basic research for the significance and value of miR-93 in rectal cancer angiogenesis evaluate.Methods:Thirty-two patients diagnosed as rectal cancer by post-operation pathology were selected randomly from Apr.2018 to Apr.2019.All patients were examined by rectal DCE-MR,K^(trans),K^(ep),V_(e),and iAUC of rectal cancer interested areas were detected.VEGF or MVD in rectal cancer tissues were detected with Western blot assay or immunohistochemistry.Expression of miR-93 in tumor or adjacent tissues was detected by qRT-PCR.Pearson rank correlation analysis was used to observe the correlation between miR-93 and tumor diameter,angiogenesis dates in cancer tissues(VEGF and MVD),MR non-morphological features(ADC and TIC type),MRI blood perfusion dates(K^(trans),K^(ep),V_(e),iAUC).Results:Expression of miR-93 in rectal cancer tissues was lower than in adjacent rectal tissues(t=9.276,P<0.001).Pearson rank correlation analysis showed that miR-93 was negatively correlated with rectal cancer diameter(r=-0.604,P<0.001,95%CI:-0.787~-0.323),negatively correlated with VEGF protein(r=-0.742,P<0.0001,95%CI:-0.867~-0.531),negatively correlated with MVD(r=-0.849,P<0.0001,95%CI:-0.923~-0.712),positively correlated with ADC value(r=0.495,P<0.01,95%CI:0.176~0.719),negatively correlated with K^(trans) value(r=-0.656,P<0.0001,95%CI:-0.818~-0.399),negatively correlated with K^(ep) value(r=-0.399,P<0.05,95%CI:-0.656~-0.058).There were no correlation between miR-93 and TIC type,V_(e) value and iAUC value.Conclusion:The expression of miR-93 was correlated with angiogenesis in rectal cancer tissues and maybe used to evaluate tumor angiogenesis in rectal cancer and to be a biological behavior prognosis marker for rectal cancer.
