丹皮酚对VEGF和SDF-1在糖尿病大鼠视网膜上表达的干预作用 被引量:4

摘要 目的探讨丹皮酚对糖尿病大鼠视网膜基质细胞衍生因子-1(SDF-1)和血管内皮生长因子(VEGF)表达的干预作用。方法将36只雄性SD大鼠随机分为正常对照组(CON组)、丹皮酚组(DPF组)和糖尿病组(DM组),每组12只。按干预时长不同,各组又再分为1月组、3月组和5月组,每组4只。通过腹腔注射链脲佐菌素建立糖尿病大鼠模型。应用实时荧光定量聚合酶链式反应(RT-qPCR)法检测各组SDF-1和VEGF的mRNA表达水平。通过HE染色观察血管内皮细胞核突破内界膜的数量。结果RT-qPCR检测结果显示,在干预1个月、3个月和5个月时间点,DM组SDF-1 mRNA和VEGF mRNA的表达水平均显著高于CON组和DPF组(P<0.05),DPF组表达水平稍高于CON组,但差异无统计学意义(P>0.05)。HE染色结果显示,DM组和DPF组大鼠视网膜突破内界膜的细胞核数均较CON组显著增加(P<0.05)。DM组和DPF组视网膜突破内界膜的细胞核数随干预时间的延长而增加,且DPF组各时间点的细胞核数均显著少于DM组(P<0.05)。结论丹皮酚可以通过下调糖尿病大鼠视网膜SDF-1和VEGF的表达来抑制视网膜新生血管形成。 Objective To explore the interventional effect of paeonol on the expressions of retinal stromal cell derived factor-1(SDF-1)and vascular endothelial growth factor(VEGF)of diabetic rats.Methods Thirty-six male Sprague Dawley(SD)rats were randomly divided into normal control group(CON group),paeonol group(DPF group)and diabetes mellitus group(DM group),with 12 rats in each group.According to the length of intervention,each group was further divided into 1-month group,3-month group and 5-month group,with 4 rats in each group.The diabetic rat model was established by intraperitoneal injection of streptozotocin.Real-time fluorescence quantitative polymerase chain reaction(RT-qPCR)was used to detect the mRNA expression levels of SDF-1 and VEGF in each group.Hematoxylin-eosin(HE)staining was used to observe the number of endothelial cell nuclei breaking through the inner limiting membrane.Results The detection results of RT-qPCR showed that the expression levels of SDF-1 mRNA and VEGF mRNA in the DM group were significantly higher than those in the CON group and the DPF group at the time points of 1-month,3-month and 5-month after intervention(P<0.05),and the expression levels of DPF group were slightly higher than those of the CON group,but the differences were not statistically significant(P>0.05).The results of HE staining showed that the number of nuclei breaking through the inner limiting membrane of the retina in the DM and DPF groups was significantly increased compared with that in the CON group(P<0.05).The number of nuclei breaking through the inner limiting membrane of the retina in the DM group and the DPF group increased with the extension of the intervention time,and the number of the nuclei in the DPF group at each time point was significantly less than that in the DM group(P<0.05).Conclusion Paeonol can inhibit retinal neovascularization by down-regulating the expressions of SDF-1 and VEGF in the retinas of diabetic rats.
