摘要
目的:建立黄芪药材和饮片的高效液相色谱-二极管阵列检测器-蒸发光散射检测器联用法(HPLC-DAD-ELSD)指纹图谱分析方法。方法:采用Agilent Zorbax SB C18(150 mm×4.6 mm,3.5μm)色谱柱;流动相为乙腈-0.5%甲酸,梯度洗脱,流速1.0 mL·min^(-1),柱温35℃,检测波长254 nm,载气气压241 kPa,漂移管温度90℃,增益50,并使用“中药色谱指纹图谱相似度评价系统”(2004A版)软件计算40批不同产地黄芪药材和饮片的相似度。结果:建立了黄芪药材和饮片的HPLC-DAD-ELSD指纹图谱方法,确立了HPLC-DAD指纹图谱的12个共有峰,HPLC-ELSD指纹图谱的11个共有峰,指认了其中的毛蕊异黄酮葡萄糖苷、芒柄花苷等6个峰。除少数几批外,黄芪药材和饮片相似度均大于0.9。结论:该方法前处理方法简单,重复性好,可用于评价和控制不同产地的黄芪药材和饮片质量。
Objective:To establish a high performance liquid chromatograph-diode array detector-evaporative light scattering detector(HPLC-DAD-ELSD)fingerprint analysis method for Astragalus membranaceus and decoction tablets.Methods:Agilent Zorbax SB C18(150 mm×4.6 mm,3.5μm)column was used;the mobile phase was acetonitrile-0.5%formic acid,gradient elution,the flow rate was 1.0 mL·min^(-1),the column temperature was 35℃,the detection wavelength was 254 nm,the carrier gas pressure was 241 kPa,the drift tube temperature was 90℃,and the gain was 50.The similarity of 40 batches of Astragali Radix and the pieces of different origins is calculated using the″chromatographic fingerprint similarity evaluation system of traditional Chinese medicine″2004A.Results:The HPLC-DAD-ELSD tandem fingerprint method for Astragali Radix and decoction pieces was established.Twelve common peaks of HPLC-DAD fingerprints were established,and 11 common peaks of HPLC-ELSD fingerprints were established,and 6 peaks such as flavonoids and formononetin were identified.Except for a few batches,the similarity of Astragali Radix and decoction pieces was greater than 0.9.Conclusion:This method has simple pretreatment method and good reproducibility.It can be used for evaluation and control of the quality of Astragali Radix and decoction pieces from different places.
