摘要
为建立比较系统科学的鸡体液或组织游离氨基酸(FAA)检测前处理方法,本试验以鸡5种体液或组织(胸肌、腿肌、血清、肝脏组织、肠道组织)为样品,经过预试调整后提出适用于鸡不同体液的前处理方法,再选取3种鸡体液或组织(胸肌、血清、肝脏)分别利用几种蛋白沉淀剂(有机盐、乙醇、有机酸)进行前处理,检测其中FAA的含量,确定除杂的适用性。结果表明:蛋白沉淀剂的选用应排除盐析,且有机酸优于乙醇。因此,检测鸡不同体液或组织样品需采用不同的前处理方法,其中,使用磺基水杨酸(SSA)纯化样品,超声波粉碎匀浆较为理想。
In order to establish systematic and scientific methods for FAA testing of chicken body fluids,5 body fluids or tissue(breast muscle,leg muscle,serum,liver tissue,intestinal tissue)of chicken were used as samples,and the pretreatment methods suitable for different body fluids of chickens were proposed after the adjustment of preliminary test.Then,three kinds of chicken body fluids or tissue(breast muscle,serum,liver tissue)were seleted to pretreat with several protein precipitants(organic salt,ethanol and organic acid)respectively,and FAA content was detected to determine the applicability of impurity removal.The results showed that the selection of protein precipitant should exclude salting out,and organic acid was better than ethanol.Therefore,different pretreatment methods were used to detect different body fluid samples of chicken,among which,purify the samples by sulfosalicylic acid(SSA)and homogenate by ultrasonic pulverization were more ideal.
